Objective: Toestablish theanimal model of type 2 diabetes millitus and to study the change of nitrotyrosine, malondialdehyde, erythrocuprein and renin-angiotensin-aldosterone system in diabete rat pancreas. And to observe the change of the oxidative stress ,renin-angiotensin-aldosterone system and insulin function intervented by Fosinopril in early stage.Methods: 21 rats which were 8 weeks old randomly were divided into three groups randomly—control group(C,n=7),diabete group(D,n=7) and Fosinopril treated group(F,n=7).C was fed with regular diets while D and F were established the animal model of type 2 diabetes mellitus by streptozotocin which was injected at a dose of 30mg/Kg body weight in abdominal.F was treated with fosinopril after the model succeeding. 8 weeks later the weight, fasting plasma glucose,fasting insuline, AngiotensinⅠ(AngⅠ), angiotensinⅡ(AngⅡ), aldosterone (ALD),MDA and SOD were measured and the expression of NT were observed by immunohistochemistry.Results: the FPG,NT,MDA,AngⅠ,AngⅡ,ALD content of D and F increased more significantly than C(P<0.05); but the body weight,FINS,IS,SOD level decreased significantly in D and F compared with C(P<0.05). The NT,MDA,AngⅠ,AngⅡcontent of F decreased than D(P<0.05), but there was no significant difference of ALD in the two groups. The body weight,FINS,IS,SOD level increased significantly in F compared with D(P<0.05).Conclusion: The level of oxidative stress and the RAAS of the rats pancreas were abormal in diabetic condition. Fosinopril could significantly inhibit the oxidative stress and decreased the RAAS, also can recuperate pancreas to some extent.
