Objective To investigate the expression and significance of 53BP1 in breast cancer ,fibroadenoma and normal mammary tissue,and analyze the association between the expression of 53BP1 and clinicopathologic category ,as well as expression of P53.Methods Immunofluorescence was utilized to evaluat the expression of 53BP1 in 20 pieces normal mammary tissues,15 samples of fibroadenoma and 48 specimens of resected breast cancer , and the correlation of this to clinicopathologic features of breast cancer was analyzed.Expression of both 53BP1 and P53 were assessed by double-labeled immunofluorescence ,and the potential association between the two was tested.Result The stable type of 53BP1 was revealed in all 20 normal mammary tissues and 15 samples of fibroadenoma. However,this type and lowDDR were excluded in 48 specimens of resected breast cancer, while ,absent expression of 53BP1 was comfirmed in 10 cases (20.8%),highDDR in 14 cases(14/48,29.2%),abnormalDDR in 24 case(s24/48,50%). Regard to the expression of 53BP1,there was no significant difference between different clinical stage of breast cancer(P>0.05) excluding 10 cases of lost type . However,significant difference was demonstrate among different pathological classification of invasive ductal breast cancer(P<0.05),and highDDR varied with pathological classification(γs=0.4955,P=0.0025),the high,the more. Additionaly,53BP1 staining was not always colocalized with p53 Expressio。Conclusion (1)DDR was absent in normal mammary tissues and fibroadenoma predominantly.(2)Absence of 53BP1 expression is proposed to be a tumor suppressor.(3) Additonally, the degree of 53BP1 focusing formation can be a potential prognostic marker of human breast cancer.
