Objective To investigate the relationship between methylation of Runx3 in promoter region and its protein expression ,and its possible relationship with genesis of carcinoma Larynx.Methods Using Immunohistochemistry technique, specimens from 55 patients of carcinoma of Larynx ( tumor tissues , adjacent normal tissues) were detected for their expression of the Runx3 gene. Meanwhile,Methylation-specific PCR was used to detect methylation of Runx3 at promoter region.Results The expression of Runx3 protein in carcinoma of Larynx were down-regulated than that in adjacent normal tissues samples (P<0.05). it was not related to clinical classification,tumor pathological grade and lymph node metastasis (P>0.05)。Methylation of Runx3 promoter was not found in adjacent normal tissues samples, but was detected in 32 cases of 55 carcinoma of Larynx. The rate of methylation of Runx3 promoter in carcinoma of Larynx was higher than that in adjacent normal tissues (P<0.05),and it was not related to clinical classification,tumor pathological grade and lymph node metastasis (P>0.05). There was correlation between methylation of Runx3 gene promoter and expression of its protein (P<0.05).Conclusion Methylation may cause the Runx3 gene inactivation in human carcinoma of Larynx. Methylation of runx3 promoter maybe correlated with genesis of carcinoma Larynx.
