ObjectiveTo study the radio-sensitizing effect of arsenic trioxide (As2O3) in human gastric carcinoma cells.MethodsAs human gastric carcinoma cells SGC-7901 selected for study.All experiments were done three times,and we took the mean as the final result.â‘ The toxic effect of As2O3 was examined to define the IC50 according to MTT assay in SGC-7901 cells,and the q value was calculated through the three group.â‘¡They were divided into four groups,such as control group,medication group,irradiation group and As2O3+ irradiation group.Cell survival rate was calculated by cloning assay,and single-hit multitarget model was used to plot the survival curve which could evaluate the effect of cell killing.Also we got Do,Dq,SER valu from it.ResultsThe IC50 in SGC-7901 cells was 12.65 umol/L. Inhibition of cell proliferation seemed more dependent on the increase of As2O3 concentration.Compared to medication group and irradiation group, As2O3+ irradiation group could significantly inhibit the cell proliferation of SGC-7901 cells.And the result demonstrated the synergism effect between As2O3 and irradiation when they were both used to kill the cells.The Do and Dq value were 2.75Gy and 2.52Gy in irradiation group.When As2O3 was administrated at 2 umo/L and 5 umol/L with irradiation,their Do,Dq,SER(Do) and SER(Dq) were 2.18Gy,1.89Gy,1.26,1.33 and 1.78Gy,1.72Gy,1.54,1.51.ConclusionsAs2O3 was shown as possessing a radiosensitizing effect in vitro in SGC-7901 gastric carcinoma cells according MTT assay and colony-forming assay. So the study was supposed to provide experimental foundation that arsenic trioxide as a radiosensitizer could be used for the therapeutic alliance of gastric cancer.
