Objective:To study the angiogenic effects of adenovirus-mediated human vascular endothelial growth factor 165 (Ad-VEGF165) taken by embolus on thromboangiitis obliterans of rat models.Methods:Totally 72 male SD rats were divided into 6 groups at random. normal group, thromboangiitis obliterans model group(TAO group), embolus group (embolus without Ad-VEGF165 femoral arterial infusion), Ad-VEGF165 vessel infusion group(Ad-VEGF165 femoral arterial infusion), Ad-VEGF165 muscle injectd group (Ad-VEGF165 intramuscularly injectd),Ad-VEGF165 taken by embolus group (femoral arterial infusion).Rat models of thromboangiitis obliterans were induced by infusing lauric acid in femoral arterial.Ad-VEGF165 taken by embolus were infused in femoral arterial. Immunohistochemistry was performed to illustrate the angiogenic effects on rat ischemic muscles after gene transferring after different interventions.Results:1. There was significant difference in the MVD marked by CD34 between Ad-VEGF165 taken by embolus group and model group(F=8.21 P <0.05),and there was also significant difference between Ad-VEGF165 muscle injectd group and model group 7 days after transfection(F=6.06 P <0.05),there was no significant difference between Ad-VEGF165 taken by embolus group to Ad-VEGF165 muscle injectd group 7 days after transfection but 14 days after transfection,the number of Ad-VEGF165 taken by embolus group was significandy higher than Ad-VEGF165 muscle injectd group(F=6.00 P <0.05).2. There was significant difference in the ratio ofα-SMA+ vessels to muscle fiber between Ad-VEGF165 taken by embolus group and model group(F=20.66 P <0.05), and there was also significant difference between Ad-VEGF165 muscle injectd group and model group 7 days after transfection(F=15.04 P <0.05),there was no significant difference between Ad-VEGF165 taken by embolus group to Ad-VEGF165 muscle injectd group 7 days after transfection ,while 14 days after transfection,the number of Ad-VEGF165 taken by embolus group was significandy higher than Ad-VEGF165 muscle injectd group(F=6.92 P <0.05).Conclusion:1:Ad-VEGFI65 can promote the neovascularization in rat model of thromboangiitis obliterans.2:The angiogenic effect is more significant in the Ad-VEGF165 taken by embolus group than another group.
