Objective:To investigate volatile oils from Amomum Villosum (VOA)'s effects on peptic ulcer recurrence through animal experiment,meanwhile the Gastric Mucosal Surface Hydrophobicity and the expression of PS2 and PAF in gastric mucosa were determined to reveal the mechanismsof VOA's effects on ulcer recurrence from the view point of the Gastric Mucosal Surface Hydrophobicity.Methods : 108 rats were randomly divided into fivegroups: normal group (n=12),sham operated group (n=12),model group(n=36), Ranitidine group (n=24) and VOA group (n=24). All rats were induced ulcer by the method of Okabe acetic squash method with some modification except for normal group and sham operated group. Rats of VOA and Ranitidine group were intragastric administrated VOA physic liquor (125mg/kg) and Omeprazole physic liquor (1.35mg/lml/100g) respectively, one time perday, totally 90 days. The others were intragastric administrated distilled water. At 90 st day, rats of modelgroup were randomly divided into model group and model recurrence group. Gastric ulcer recurrence model was duplicated by IL-1β(1μg/kg)intraperitoneal injection except for rats of model group which were intraperitoneal injection saline instead,At 7th, 92th day (92 th dayis also 48h later after ulcer recurrence was induced),12 rats of each group were put to death and the stomach was taken out.After macroscopic observation of stomach and measurement of gastric ulcer index(GUI), the gastric barrier mucus were used to detect the content of hexosamine and phospholipid,the gastric tissue which contain ulcer(draw the materials from correspondingsites of rats without ulcer) was obtained for histological investigation by HE staining,PS2,PAF and protein detection by immunohistochemistry.Results:①At the 7th day after model was made,the ulcer foci in rats were observed macroscopically and histologically at different degree. GUI of model group is the highest, while the GUI of VOA group and Ranitidine group were both significantly lower than that of model group(P<0.01).At the 92th day, compared with recurrence group, GUI of Ranitidine group and VOA group were lower(P<0.01), while the CA of the two group were higher (P<0.01).②At the 7th day, the content of hexosamine and phospholipid of recurrence group and Ranitidine group were both significantly lower than that of normal group (P<0.01). At the 92th day, the content of hexosamine and phospholipid of VOA group and Ranitidine group were both significantly higher than that of recurrence group (P<0.01), and there was significant difference between the two (P<0.01). There was no difference between VOA group with the normal group.③At the 7th day, The expression level of PS2 in normal group and sham operated group were lowest. The expression level of PS2 in VOA group were higher than recurrence group (P<0.01), while Ranitidine group was not(P>0.05). At the 92th day, the expression level of PS2 in Ranitidine group and VOA group were higher than recurrence group (P<0.01), and there were significant differences between them (P<0.01).④PAF protein expressed less in normal group and sham operated group while there were significant more expression in Ranitidine group and model group at 8th day. Villosum volatile oil can significantly reduce the expression of PS2 of the gastric tissue. At the 92th day, the expression level of PAF in Ranitidine group and VOA group were both lower than recurrence group (P<0.01), but Ranitidine group was higher than normal group (P<0.01), and there was no significant difference between VOA group with normal group (P>0.05).⑤The expression of PS2 had a positive correlation with both the content of hexosamine and phospholipid (P<0.01). The expression of PAF had a negative correlation with both the content of hexosamine and phospholipid (P<0.01).Conclusion:①Through the monitoring of these target, it is validated by experiments that using the glacial acetic acid ulcer model reliable and IL-1βrelapse model of success.②Compared with the Ranitidine, volatile oil villosum could better treat gastric ulcer of rats and to prevent its recurrence.③VOA could inhibit activation of PS2, down-regulated the over-expression of PAF, also by affecting gastric the content of hexosamine and phospholipid, thus improved the Gastric Mucosal Surface Hydrophobicity effect, which maybe one of mechanisms of VOA's anti-relapse effect.
