Objective:To study the expression of pRb2/p130 protein and CDK4 protein in endometrioid adenocarcinoma and their relationship with Pathological clinical features. To explore the relationship between the expression of pRb2/p130 and CDK4 and evaluate its possible significant role in tumorigenesis and progression of endometrioid adenocarcinoma.Material and Method:A immunohistochemical method of Streptavidin peroxidase was used to detect pRb2/p130 protein and CDK4 protein in 28 cases of normal endometrium tissues ,20 cases of atypical hyperplasia of endometrium, and 30 cases endometrioid adenocarcinoma.To analyze the relationship to age,clinicalstage,histolgical grade, muscular invasion,and lymphnode metastasis.Results:1.The expression of pRb2/p130 proteinThe expression of pRb2/p130 was gradually reduced in the normal endometrium,endometrial atypicall hyperplasia and endometrial adenocarcinomas. (Followed by 82.14﹪,55.00﹪,23.33﹪),And among the three groups have a statistical meaning(P<0.05), and the two among them also have a significant statistical meaning.The expression of pRb2/p130 protein in endometrial atypicall hyperplasia and endometrial adenocarcinomas were lower than that in the normal(P<0.05). The expression of pRb2/p130 was not correlated with the age of patients, clinicalstage , muscular invasion and lymphnode metastasis(P>0.05). but was negatively correlated with the histolgical grade(P<0.05), the worse differentiation,the lower pRb2/p130-positive rate(P<0.05).2.The expression of CDK4 proteinThere was obvious difference of CDK4 positive rate among in the normal endometrium,endometrial atypicall hyperplasia and endometrial adenocarcinomas (Followed by 40.00﹪,60.00﹪,86.67﹪), And Among the three groups have a statistical meaning(P<0.05), and the two among them also have a statistical meaning. The expression of pRb2/p130 protein in endometrial atypicall hyperplasia and endometrial adenocarcinomas were higher than that in the normal(P<0.05).The expression of CDK4 was not related to the age of patients , clinicalstage, histolgical grade and lymphnode metastasis(P>0.05). but was related to muscular invasion degree(P<0.05). the worse depth of invasion,the higher CDK4-positive rate(P<0.05).3.The relationship between the expression of pRb2/p130 protein and CDK4 protein in endometrial adenocarcinomas.When the positive of pRb2/p130 protein expression was 7 cases, the positive of CDK4 protein expression for 4 cases in 30 cases of endometrial adenocarcinomas.The positive rates for 57.1%.When the negative of pRb2/p130 expression was 23 cases,the positive of CDK4 protein expression for 22 cases in 30 cases of endometrial adenocarcinomas.The positive rates for 95.7%.Using Spearman rank correlation analysis to analyse the expression of pRb2/p130 protein and CDK4 protein in endometrial adenocarcinomas in 30 cases.There was a negatively correlation between the expression of pRb2/p130 and that of CDK4(rs′=-0.4791,P=0.0307).Conclusion:1. According to the phenomenon of the positive rate of pRb2/p130 protein expression were gradually reduced in the normal endometrium,endometrial atypical hyperplasia and low differentiated endometrial adenocarcinomas,it was showed pRb2/p130 play a negatively regulation role in endometrial carcinoma.2.From the normal endometrium,through endometrial atypical hyperplasea ,to endometrial adenocarcinoma, the positive rates of CDK4 protein expression were gradually increased ,and it was related to the invasion degree,but was not related to the age of patients,clinicalstage, histological grade and lymphnode metastasis.3.There is negative correlation to the expression of pRb2/p130 protein and CDK4 protein in endometrial adenocarcinomas.4.The pRb2/p130 protein expression decline or loss and abnormal expression of CDK4 protein in endometrial adenocarcinomas might play synergetic roles in carcinogenesis and evolution of endometrial adenocarcinomas.
