BACKGROUND: Breast cancer is one of the most common malignant tumors in women.NAC has become more important in combined therapy of breast cancer,especially the Locally advanced breast cancer.The selection of a systemic breast cancer therapy is based on the expression pattern of prognostic markers,which would be altered by the primary systemic therapy. In our study we sought to determine whether neoadjuvant chemotherapy may alter these expression patterns within the tumors.OBJECTIVE: The main purpose of this article is to explore the level of protein molecules in the neoadjuvant chemotherapy for breast cancer molecular biomarkers ER, Her-2 impact. Help to achieve the choice of breast cancer treatment and prognosis purposesMETHODS: Hollow needle aspiration before chemotherapy, after chemotherapy in patients with surgical resection of tumor tissue samples, frozen to preserve. Extraction of protein, using protein electrophoresis, western blotting detected 27 cases of breast cancer patients (patients with non-PCR) of the preoperative and postoperative tumor ER, Her2-coded by level of protein expression were observed before and after neoadjuvant chemotherapy for protein expression changes. The results of an electronic file form, unity is converted to tif format, using "Quantity One" analysis software for processing, the protein expression in various bands of the average density values .The results : 27 cases of neoadjuvant chemotherapy in breast cancer cases: ER expression in 10/27, no significant changes in 5 / 27, increased expression of 12/27. Chemotherapy before and after the change was not significant (p = 0.397); Her-2 expression decreased 15/27, no significant changes in 3 / 27, increased expression of 9 / 27. chemotherapy before and after changes in the expression of Her-2 were statistically significant (p = 0.044)CONCLUSION: The Her-2 and ER gene expression changes in the protein level in patients with breast cancer before and after NAC, ER protein expression were higher. The possible reason is the existence of the entire tumor tissue within the biological differences or may be caused as a result of sample variation. Her2 protein get lower after the neo-adjuvant chemotherapy. May be the Her2 in patients with high expression of tumor cells with high proliferative activity, more sensitive to chemotherapeutic drugs, resulting in protein expression after chemotherapy reduction. Chemotherapy in patients with a longer cycle of its Her2 protein expression were higher than the short cycle of chemotherapy in patients with low, this may be due to high expression of Her2 on the tumor cells with doxorubicin doses of chemotherapy dose-effect relationship...
