Th1/Th2 Cells Immunomodulatory Effects Of EGCG And PS-T On Non Small Cell Lung Cancer

OBJECTIVE: To investigate the immunomodulatory effect of EGCG and PS-T on non small cell lung cancer's Th1/Th2 cells immune response state.METHODS: (1)10 normal subjects served as controls, 15 benign disease patient served as controls,25 patients suffered from III-IV stage non small cell lung cancer were studied. (2)To separate human peripheral blood lymphocytes.(3)To cultivate lymphocyte and interfere by EGCG and PS-T.(4)Supernatant concentrations of Th1 cytokines (IFN- r)and Th2 cytokines (IL-4) were assayed by enzyme-linked immunosorbent assay(ELISA).(5)Proportion of Th1 cells and Th2 cells were assayed by immunocytochemistry method.(6)The correlations between IFN-r/Th1, IL-4/Th2 with EGCG and PS-T's effect were analyzed respectively.RESULTS: It was found that patients with non small cell lung cancer had higher IL -4/Th2 levels compared to patients with benign disease and normal subjects (P<0.01), whereas having lower IFN-r/Th1 levels compared to patients with benign disease and normal subjects(P<0.01),and there weren't statistical differences in the levels of IFN- r/Th1 and IL-4/Th2 between patients with benign disease with normal subjects(P>0.05). We noticed that PS-T group and EGCG group had higher IFN-r/Th1 levels compared to lung cancer without intervention group (P<0.01), whereas having lower IL-4/Th2 levels compared to lung cancer without intervention group(P<0.01). And two medicines combination group had higher IFN-r/Th1 levels compared to PS-T group and EGCG group(P<0.05), whereas having lower IL-4/Th2 levels compared to PS-T group and EGCG group(P<0.05). In PS-T group, IFN-r level was positively associated with PS-T dose (R=0.294,P<0.01), whereas IL-4 level was negatively associated with PS-T dose (R=-0.308,P<0.01) . And in EGCG group, IFN-r level was positively associated with EGCG dose (R=0.307,P<0.01) , whereas IL-4 level was negatively associated with EGCG dose (R=-0.275,P<0.01) .CONCLUSION: A Th2 pattern of immune response of characteristic of cellular immunity is dominant in non small cell lung cancer, PS-T and EGCG have positive immunomodulatory effect on non small cell lung cancer' Th1 cytokine, also can inhibit Th2 cytokines respectively, and the effect is dependent on dose. Combination of two medicines had addition effect.
