Objective:1.To investigate the impact of ischemic postconditioning on hepatocytic apoptosis and to provide experimental evidence of the protection of liver function after clinic partial hemi-extracorporeal hepatectomy.2.To investigate the potential relationship between ischemic postconditioning and the expressions of Bcl-2 protein.3.To investigate the impact of Bcl-2 on hepatocytic apoptosis.Methods:The research targets are healthy male SD rats.1.Experimental animals were randomly divided into 3 groups and each group has 40 rats.The groups are:supposititious-operation(S) group, ischemia-reperfusion(IR) group,ischemic postconditioning(IPO) group. Establish partial hemi-extracorporeal hepatectomy model of rats in IR and IPO group.Then resect left partial liver after cold preservation at 0-4℃. Ischemic postconditioning is actualized in IPO group before reperfusion.2.Blood and liver tissues were harvested at Oh,0.5h,1h,2h,4h,8h,12h and 24h after reperfusion in each group:①measure the level of alanine aminotransferase(ALT);②measure the hepatocytic apoptosis;③measure the expressions of Bcl-2 protein.3.All the experimental data was analysised by statistic method.Result:1.The level of alanine aminotransferase(ALT) of rats in each group was heightened at different degree and declined subsequently.Compared with other groups,the peak value of ALT in IPO group was significantly lower(P<0.05).2.Compared with S group,the hepatocytic apoptosis index of liver tissues was significantly higher(P<0.05) both in IPO group and IR group.The HAI of IPO group was significantly lower(P<0.05) than one of IR group.3.The expressions of Bcl-2 protein after reperfusion in S group was few.Compared with IR group,the expressions of Bcl-2 protein of IPO group was significantly higher(P<0.05).4.The expressions of Bcl-2 protein both of IR group and IPO group fell after reperfusion,while the hepatocytic apoptosis rised.The Bcl-2 protein A value and the corresponding HAI is minus-related in both group(IR group r=-0.936,P=0.001;IPO group r=-0.912,P=0.002).Conclusion:1.The incidence of hepatocytic apoptosis after partial hemi-extracorporeal hepatectomy can be reduced by ischemic postconditioning.2.The increase of the expressions of Bcl-2 protein is one of the potential mechanism of ischemic postconditioning in reducing the hepatocytic apoptosis.3.High expressions of Bcl-2 protein closely related to hepatocytic apoptosis.
