Objective:Utilize the mice zona pellucid-3(pzp3) polytide to set up the animal model of autoimmune premature ovarian failure(POF) and discuss the impact of glucocorticoid on experimental autoimmune POF mice in different time.Method:The 70 mice were randomly divided into the control group,the early,the medium,the late glucocorticoid treatment group and the early,the medium,the late model group,10 in each group.The control group mice were injected with distilled water in the sole while the rest mice were injected with CFA-PZP3 mixed solution which contained CFA 0.05ml and pZP3 50nmol to set up the animal model of autoimmune POF. The early,the medium-time,the late treatment groups begin to treat with glucocorticoid by intraperitoneal injection separately on the 10th,15th,20th day from CFA-PZP3 inject day,and the model groups were treated with distilled water at the same time with their corresponding treatment group. They were all treated and observed 3 sex cycles.The observed standards were:1.The change of sex cycle;2.Detection of AzpAb serum level by ELISA;3.Observation of the histological changes of the mice by HE slicing under optical microscope;4.Evaluation of the severity of autoimmune oophoritis by examining the infiltration of CD45 expressing in the ovary.Result:①The model group's sex cycle disorder rate/the AzpAb level in serus/the expression rate of CD45 seperately were 90%,>32 ug/L 1,90%,obviously higher than the control group which corresponding value were 10%,17.55 ug/L,30%;the growing follicle rate in model groups were 22.44%-27.74%,obviously lower than the control group which value were 42.59%,the difference all had statistical significance,p<0.05.②The medium-time treatment group's sex cycle disorder rate/the AzpAb level in serus/the expression rate of CD45 seperately were 30%,18.67 ug/L,10%,obviously lower than its model group whose value were 100%,33.05 ug/L,90%;the growing follicle rate in the medium-time treatment group was 41.35%,obvously higher than its model group whose value was 23.7%,the difference all had statistical significance,p<0.05.③The early,the medium,the late treatment group's AzpAb level in serus seperately were 27.22 ug/L,18.67 ug/L,27.04 ug/L,the medium treatment group were obviously lower than the rest groups;their growing follicle rate seperately were 30.53%,41.35%,33.64%,the medium treatment group were obviously higher than the rest groups,the difference all had statistical significance,p<0.05;the early and late treatment group had no statistical significance comparison with each other,P>0.05.Conclusion:1.utilize pzp3 sole injection can successfully set up the animal model of autoimmune POF.2.glucocorticoid can treat the autoimmune POF effectly.3.glucocorticoid treat the autoimmune POF mice the most effective in medium-time.
