| Objective: To investigate the significance and change of expression of CHOP after acute spinal cord injury and explore the contribution of ERS to the spinal cord injury.Methods: Rat model of ASCI was established with a modified Allen's method which located in the T8-T10.The experimental rats were randomly divided into two groups: group test performed with SCI; group sham with false operation. The animals were sacrificed at 3h, 24h, 3d, 7d respectively after injury. The nerve function of spinal cord was evaluated by modified Tarlov score. The morphologic character was used HE. The expression of CHOP were detected by immunohistochemistry method. Cell apoptosis was examined using the TUNEL assay .The expression of CHOP and apoptosis was measured by use of IPP (Image-Pro Plus 6.0) .The result was showed by IOD(integrated optical density) and density (mean).Results: The number of paralysis rats were not significant in the group sham and the group test was observed with the different severities of the Motor Deficit. The Tarlov scores of and group sham were higher than the group test (P<0.01) . Necrosis cells and apoptotic cells were not significant in the group sham but were noted in rats after SCI. The expression of CHOP of group test was higher than the group sham at different times (P<0.01) and it began at 1d, peaked at 3d and decreased at 7d after SCI. Apoptotic cells in the group test was consistent with the expression of CHOP which increased at 1d, peaked at 3d day and decreased at 7d after SCI and was 3h<1d<3d>7d(p<0.01). The expression of CHOP was related with the apoptosis and the coefficient correlation (Preason's) is 0.644.Conclusion: ASCI could induce the expression of CHOP by means of the ERS, and this was related to the apoptosis of neurocyte. |
