Objective:To investigate the expression of ICAM-5 in oxygen-glucosedeprivation neuronal modelMethods:1.To establish the OGD neuronal model and make verification.2.To use PAJU-ICAM-5 cell line which express normal ICAM-5 and PAJU-NEO cell line which express neo resistant as neuronal models.Both PAJU-ICAM-5 and PAJU-NEO cells were treated with OGD in the same times respectively.Then,we performed western blot for detecting expression of ICAM-5.Flow cytometer(FCM) was used to exam apoptosis of both treated PAJU-ICAM-5 and PAJU-NEO cells and phase contrast microscope was used to observe morphology of OGD treated PAJU-ICAM-5 and PAJU-NEO cells.Meanwhile the lengths of cells process were micro-measured.Results:1.The OGD model was established and verified by expression of hypoxia inducible factor-1a(HIF-1a).HIF-1aexpression was detected by immunofluorescence and western blot.2.Long time expose to oxygen-glucose deprivation reduce expression level of ICAM-5.3.The apoptosis rate of PAJU-ICAM-5 cells is lower than that of PAJU-NEO cells after both cell lines were treated with OGD in the same time.Morphologic injuries of PAJU-ICAM-5 cells are also light compare to that of PAJU-NEO cells.Length of PAJU-ICAM-5 cell processes are longer compare to that of PAJU-NEO cells. Conclusions:1.Long time expose to oxygen-glucose deprivation may reduce expression level of ICAM-5.2.Expression of ICAM-5 can protect neurons from oxygen-glucose deprivation injury.
