Objectives1.To investigate the correlation between polymorphism of vitamin D receptor(VDR) Fok I restriction endonuclease site and the susceptivity of gastric cancer in Han People of Shandong Province.2.To evaluate the role which vitamin D receptor Fok I restriction endonuclease polymorphism had played to the pathological differentiation and AJCC staging of gastric cancer patients after surgery.MethodsA case-control study method was adopted.40 gastric cancer patients who came to Shandong Provincial Hospital from July,2008 to March 2009 and 109 healthy control persons were enrolled in this study to analysis the correlation between polymorphism of vitamin D receptor(VDR) Fok I restriction endonuclease site and the susceptivity of gastric cancer in Han People of Shandong Province.The clinical base-line data were collected using a structured questionnaire containing the subjects' clinical characteristics,including age,gender,smoking,drinking,family history of gastric cancer,pathology postoperative.DNA of VDR genotypes were determined by Fok I restriction endonuclease digestion of Polymerase Chain Reaction—Restriction Fragment Length Polymorphism(PCR-RFLP).Statistical analysis were performed using SPSS software-version 11.5.Comparison between two groups was performed using the chi-square test,unpaired t-test or non-parametric means test for continuous variables.After adjustment for confounding factors,a multivariate logistical regression analysis was performed to identify the odds radio and 95%CI.A P-value of<0.05 was considered statistically significant. Results1.Hardy Weinberg equilibrium was evident for the gene polymorphism of VDR-Fok I restriction endonuclease in both case group and control group.2.The F allele frequencies in healthy persons and in gastric tumors patients were 61.5%and 42.5%respectively,which had a statistical difference(P<0.05).A multi-variable logistic regression showed the f allel was susceptibility allel of gastric cancer(OR=2.051,95%CI=1.220~3.447).3.The genotype frequencies of FF,Ff and ff in healthy persons and in gastric tumors patient were 39.5%,44.0%,16.5%and17.5%,52.5%,30%respectively, with statistical differences(P<0.05).The ff genotype was more frequently identified in patients with poorly pathological differentiation(P<0.05),however, the genotype frequencies of FF,Ff and ff in different AJCC stage groups showed no statistical difference(P>0.05).Conclusions1.VDR Fok I polymorphisms was associated with the incidence of gastric cancer in Han people of Shandong Province,and f allel might be the susceptibility allel of gastric cancer.2.VDR Fok I polymorphisms was associated with the pathological differentiation of gastric cancer,which suggested that f allel might be linked to a subset of gastric cancer showing specific biological behaviors.
