As the result, side branches of Glycyrrhiza 3~5 years old is chosen for it high content of liquorice acid and Glycyrrhiza polysaccharides.In distilling technology study, single-factor and orthogonal experiments were designed. The content of Glycyrrhiza polysaccharides was measured by phenol-vitriol method while glucose was used as comparison. And the best abstracting conditions are 90°C, 0.5 hours for twice and 30 times liquid divides stuff.In the purification process of Glycyrrhiza polysaccharides, best techniques of ethanol deposit was ascertain by orthogonal experiments, and the concentration of ethanol was 80 % at RT for 12 hours. Sevage method was considered as the best way of separating the protein impurity in Glycyrrhiza polysaccharides. The Glycyrrhiza polysaccharides freezing dry without any small molecule impurities and protein impurities were called GPS in the following text. Certain concentration solutions were confected by dissolving the GPS with 0.9% NaCl solution. Feed the solution to BalB/C to study the immune-modulation in vivo. Carbon grains expurgation experiment, lymphocyte traform experiment and hemolysin in blood serum experiment were the main bodied of these part. It was shown that GPS can enhance the differential immunity capacity and non differential immunity capacity obviously (P<0.01) .After dissolving the GPS in the distilled water again, and separating by DEAE-cellulose and sephadex G-100 gel chromatogram columniations, four different kinds of Glycyrrhiza polysaccharides were obtained and named as GPS-1b, GPS-2a, GPS-3a and GPS-4a. And then GPS-2a, GPS-3a and GPS-4a.were collected as the samples for characteristic structure analyzing and study on immune-modulation. They were ascertained to be polysaccharides by IR spectroscopic analysis. Their total sugar content was 87.8 %, 86.8 % and 82.3 % measured by phenol-vitriol method while the protein content was 1.88 %, 7.54 % and 9.89 % measured by Bradford method. The sugar components were ceanothus sugar, Arabia sugar, xylose, galactose and glucose. The pictures of AFM observing results showed differences in microcosmic configurations. They appeared as plane reticulations, solid reticulations and disperse congeries in different sizes.
