Ardisia gigantifolia Stapf of Myrsinaceae genera,which is one of traditional Chinese herbal medicine,plays a role in invigorating blood circulation, eliminating stasis,etc.And it used to treat the wounded after a fall in the area of Yao minority in Southern China commonly.At present,experimental studies of Ardisia gigantifolia Stapf are rarely reported,especially,effect on thrombosis,there are fewer reports in domestic and abroad.Therefore,this article was related to the followings.Trial 1 Extraction and Content Determination of plysaccharides,saponins and alkaloids of Ardisia gigantifolia Stap.In this article,total polysaccharides,total saponlns and total alkaloids were determinde by UV-Vis spectrophotometry.Glucose was used as a reference substance,the total polysaccharides calculated in glucose(C6H12O6) in extract of Ardisia gigantifolia Stap is about 2.30%.Ginsenoside Rg1 was used as a reference substance,the total saponins calculated in ginsenoside Rg1(C43H74O13) in extract of Ardisia gigantifolia Stap is about 1.01%.Oxymatrine was used as a reference substance,the total alkaloids calculated in oxymatrine(C15H24N2O2) in extract of Ardisia gigantifolia Stap is about 0.28%.Trial 2 Screening of the antithrombotic activity of Ardisia gigantifolia Stapf extracts.The results show that:compared with the control group,in polysaccharide group, a significant prothrombin time(PT),activated partial thromboplastin time(APTT), prolonged in recalcification time(RT),and inhibit thrombase activities in vitro,in saponin and alkaloid groups,a significant prolonged in recalcification time(RT);When compared with the polysaccharide group,in saponin and alkaloid groups,a significant decurtation in RT,a significant attenuated in inhibit thrombase activities.In the optical density experiments,the anticoagulant activity of polysaccharide is best.Comprehensive analyzing our results,polysaccharide is regarded as the active ingredient of our further studying antithrombotic effects.The following test is in addition to screening of the antithrombotic activity of Ardisia gigantifolia Stapf extracts.Investigated effects of Ardisia gigantifolia Stapf polysaccharides on blood clotting system,hemorrheology and pulmonary injury in rat.48 Sprague-Dawley(SD) rats were randomly divided into 6 groups(n=8 in each group), including the control,patho-model,PNS and 3 Ardisia gigantifolia Stapf-treated model groups of 708 mg/kg,354 mg/kg,144 mg/kg crude drug.At 9:00,panax notoginseng saponins(PNS) and 3 Ardisia gigantifolia Stapf-treated model groups were intragastrically treated in 1mL/kg body weight(WB).Whereas the control and patho-model groups were administered with the same volume of 0.9%NaCl solution in the same way.At 10:00 in the same day,except for the control group that was injected with 0.9%NaCl solution,all others were subcutaneously injected with adrenailne 0.08 mg/kg body weight(WB) to induce blood stasis and pulmonary injury.The operations were done once a day for 12 days.After the last injecting with adrenaline,took blood for assaying of blood physiological and biochemical index,drew lung for pathological test.This results showed that there was a significant change in prothrombin time(PT),activated partial thromboplastin time(APTT), levels of hematocrit(HCT),concentrations of hemoglobin concentration(HGB).In addition,light microscopic viewing of the slides demonstrated a marked pulmonary injury in the patho-model group.When compared with the patho-model group,In the model-treated group of708mg/kg,a significant prolonged PT,thrombin time(RT) and APTT,and decrease in levels of Fg,HCT and HGB.Furthermore,HE staining showed that pulmonary injury was obviously improved in the 708mg/kg group.In short,Ardisia gigantifolia Stapf polysaccharides could significantly prolong PT,TT and APTT,decrease in levels of Fg,HCT and HGB in vivo to inhibit intrinsic and extrinsic coagulation system,thus to prevent thrombogenesis and attenuate pulmonary tissue damage.
