Despite the fact that entomogenous fungi are a small group of organisms, recent researches revealed that they can produce special bioactive compounds with unique structures and functions which may promise a good application for-future. This aroused an upsurge of research on metabolites of entomopathogenic fungi in recent years.During a screening for bioactive metabolites entomogenous fungi, the fermentation broth of a strain of Hirsutella sp. RCEF0851 exhibited strong immuno- enhancive property. Therefore, we launched a systematic study on immunomodulatory activities of different extracts of mycelia of RCEF0851 by using Real-Time-PCR method.The results showed that ethyl acetate extract can significantly increase the level of TNF-a transcription, but the values of 2-△△Ct of cytokines IL-1βand IL-6 were not changed significantly compared to DMSO control group. After cells co-culture with ethyl acetate extracts at the same concentration in different time, the ethyl acetate extracts can enhance gene transcription of TNF-α, however, the level of TNF-a transcription did not changed significantly after co-cultured 12 hours.Cells co-culture with different concentrations of water extracts showed that the values of 2-△△Ct of IL-1βincreased gradually as the concentration increased, but the values of 2-△△Ct of IL-1βwere not changed at the concentration high than 300ug/mL. When cells co-cultured with water extracts at the same concentration, the values of 2-△△Ct after cultured 12 h and 24h were higher than that of 6 hours. Water extracts can significantly increase the level of TNF-a transcription.Cells co-cultured with different concentrations of methanol extract showed that the values of 2-△△Ct of methanol extracts were not change significantly compared to DMSO control group which means the concentration of the extract did not affect the level of IL-1βgene transcription. The values of 2-△△Ct was increased significantly when the cells co-cultured more than 12 hours with methanol extracts at the same concentration. At the same time the values of 2-△△Ct of cultured after 24h were higher than that of 12 h. Methanol extracts can significantly increase the level of TNF-a transcription.
