Objective:To study the protective effect of hyperbaric oxygen early intervention on multiple traumatic rabbits,through observing the dynamic changes of the interleukin-8(IL-8). interleukin-10(IL-10) in blood plasma and the pathologic changes of heart,lung and kidney tissue.Methods:1.Animal subgroup:Rabbits(mean weight 2.15±0.15kg):animals were randomly subjected three groups.Each group has 8 rabbits.The control group:no shock no fracture;the multiple trauma group:induce shock and fracture then blood transfusion and fluid infusion for resuscitation in regular.The hyperbaric group:induce shock then blood transfusion and fluid infusion for resuscitation in regular.Post in hyperbaric oxygen chamber at once,first,denitrogenation by flushing with 100%oxygen 5 minute and maintain 0.12Mpa,5 minute later pressurize to 0.2 Mpa oxygen concentration>95%40 minutes. Decompression takes 10 minutes.2.Detection:After anesthesia is counted Oh,induce shock in 0.5h(control group does not induce,expose and isolate blood vessel only),resuscitate in 1.5h,resuscitation was complete in 2.5h.According to 0.5h,1.5h,2.5h,4.5h,8.5h,24h.,there are six subgroup in each group animals(n=8).Get 2ml blood from each animals,get red blood cell on the bottom when it's centrifuged.The rest get blood plasma when them was centrifuged.3.Histological procedures in 24h,the heart,lung,kidney were excised from each rabbit when still live,and fixed with 1%paraformaldehyde.Fixed tissues were embedded in paraffin and processed for light microscopy.Sections were stained with hematoxylin and eosin.4.Statistical analysis:The groups were compared with one way ANOVA,and the Student Newman Keuls post hoc test(using SPSS1 3 soflware).Differences were considered significant when P<0.05.Results:1.Interleukin-8(IL-8)of Blood plasma:The difference is no significant,from which IL-8 of blood plasma in three groups in 0.5h.The diference between HBO group and multiple trauma group is no significant in1.5h,2.5h,4.5h,the difference between control group and multiple trauma group was significant P<0.05,the difference between control group and HBO group was significant P<0.05.The quantity of IL-8 from animal's blood of hyperbaric group was lower than the multiple trauma group in 8.5h,24h.The diference was significant P<0.05.The quantity of IL-8 from animal's of multiple trauma group and HBO group was increased than the control group.The diference was significantP<0.05.2.Interleukin-10(IL-10)of Blood plasma:The difference is no significant,from which IL-10 of blood plasma in three groups in 0.5h,1.5h.The diference between HBO group and multiple trauma group is no significant in 2.5h,4.5h.The difference between control group and multiple trauma group was significant P<0.05,the difference between control group and HBO group was significant P<0.05.The quantity of IL-10 from animal's blood of hyperbaric group was lower than the multiple trauma group in 8.5h,24h. The diference was significant P<0.05.The quantity of IL-10 from animal's of multiple trauma group and HBO group increased than the control group.The diference was significant P<0.05.3.Pathologic microscopic examination:In 24 hours later,killed the animals and cut heart,lung,kidney then observed the difference in light microscope.In multiple trauma group, the pathologic lesions of the organs were very obvious.But in HBO group,there were less pathologic lesions of the organs,and in control group,there weren't obviously pathologic lesions of the organs.Conclusion:Early intervention HBO after resuscitation of multiple trauma can decrease the levels of plasma IL-8,IL-10 and abate inflammatory effusion, tissular edema of the heart,lung and kidney.
