[Objective]To explore the type of the joint role and possible mechanism of action on the offspring of male reproductive system after different doses ofnonylphenol and estradiol exposure during pregnancy[Methods]Selective mature,200±20g body weight of SD rats,male and female SD rats according to a 1:2 ratio of mating with the cage to get pregnant rats,and then the pregnant rats were randomly divided into 8 groups(A,control group;B,40mg/kg NP;C,80mg/kg NP;D,160mg/kg NP;E,40mg/kg NP +20 ug / kg E2;F,80mg/kg NP+20 ug / kg E2;G,160mg/kg NP+20 ug / kg E2;H,20ug/kg E2;) in pregnant rats 14-19 days pregnant,NP exposure through oral administration,estradiol exposure through intrapefitoneal injection.Detection of F1 male rats:1,using real-time fluorescence quantitative PCR detection of 19 days in the fetal rat testis expression level ERβmRNA and MISmRNA.2,birth indicators:measured the Body length,Tail length,Anogenital distance.3,Organ coefficient of prostates,testis and epididymis were measured and then testis were conserved in 10%formalin for pathological examination at the age of 90 days,the daily gonepoiesis quantity and sperm counts in the tail of the epididymis were tested through the Light microscope.The level of serum testosterone were measured by Radioimmunoassay.[Result]1,birth indicators:compared with control group,nonylphenol and estradiol shortened F1 rats from birth Anogenital distance,Body length and tail length, its role in the first two types of joint role for the add.2.F1 male rat reproductive system 90-day-old indicators:comparison with control group,each group of testis, epididymis,prostate organ coefficient,sperm activity,epididymal tail sperm count, daily testicular sperm production and serum testosterone were decline;the two factors on the coefficient of testis,prostate coefficient,sperm activity,epididymal tail sperm count,daily testicular sperm production and serum testosterone for the sum of the joint role of the type of role.3.ERβmRNA and MISmRNA expression level: nonylphenol and estradiol can elevated ERβmRNA and MISmRNA expression,the combined effects of the type of role for the add in the former.[Conclusions]1,nonylphenol and estradiol may affect the MIS,ERβsynthesis and lead to lower testosterone level,thus have an adverse impact on Male reproductive system.2,nonylphenol and estradiol in the role of the existence of a joint,the joint effect of type of additive effect...
