Objective:The purpose of the study is to investigate the role ofRunx3,TGF-β1 andβ-catenin genes in oncogenesis of colon carcinoma by detecting the methylation rate of Runx3 and protein expression of Runx3,TGF-β1 andβ-catenin in tissues of colon carcinoma,adenomatous polyposis coli and normal colonic mucosa.Methods:Methylation-specific PCR was used to detected the methylation rate in tissues of colon carcinoma(n=34),adenomatous polyposis coli(n=34) and normal colonic mucosa(n=34).The protein expressions of Runx3,TGF-β1 andβ-catenin in same specimens were also detected by immunohistochemical staining.X~2 Test and Fisher's exact Test were applied to analyze the data. Results:1.The methylation rates of Runx3 were 23.5%(8/34) for the specimen of colon carcinoma 20.6%(7/34) for that of adenomatous polyposis coli and 0%for that of normal colonic mucosa.The methylation rate of Runx3 for colon carcinoma was significantly higher than that of normal colonic mucosa(P<0.05) and no statistical difference comparing with that of adenomatous polyposis coli (P>0.05 )2.Positive protein expression of Runx3 was 17.7%(6/34) in colon cancer which was much lower than that of colonic 61.8%(21/34) and normal tissues 76.5%(26/34).Significant difference was observed between colon carcinoma and colonic polypi groups,colonic cancer and normal tissues(P<0.05).The latter two groups showed no significant difference(P>0.05).3.The positive expression of TGF-β1 was 88.3%(30/34 cases) in colonic cancer. Which was much higher compared with colonic polypi and normal colonic tissues. Significant difference were observed between colon cancer and polypi,colonic cancer and normal tissues(P<0.05).The latter two groups was significant statistically.4.The positive expression ofβ-catenin was 79.4%(27/34) in colonic cancer.Which was much higher than that of colonic polypi and normal tissues.Significant difference were observed between colonic cancer and polypi,colonic cancer and normal tissues(P<0.05).The latter two groups was not Significantly different (P>0.05).5.Correlation coefficient of TGF-β1 andβ-catenin in colonic cancer equaled to 0.223(P=0.018).But negative correlation was found between Runx3 and TGF-β1.r =-0.40(P=0.016). Conclusions:The expressions of TGF-β1,Runx3 andβ-catenin were closely related with colonic cancer,Which suggesting a possible involvement of TGF-β1,Runx3,β-catenin in carcinogenesis and progression of colonic cancer.Detecting methylation of Runx3 in colonic cancer may provide new tools for early diagnose and prevention of the disease.
