[Background]:A large number of reports that tracheal epithelial cell is one of the most important influence factors of OBS.Recently,tracheal heterotopic transplantation model could certificate that reports.[Objective]:To study the influence of tracheal epithelial cell effects to OB in tracheal heterotopic transplantation model.[Design,Time and Setting]:A controlled observational experiment was performed at Open Laboratory of Molecular Biology in Tongji University and Animal Laboratory in the Second Military Medical University,from September 2007 to July 2008.[Material]:Sixteen male SD rats and eight Wistar male rats weighing 200-250 g were included.Fetal bovine serum,trypase,PBS and DMEM(Gibco company) were used for the study.[Methods]:All rats were divided into four groups:A group,Wistar rat tracheal model(donator) subcutaneous implanted in the back of SD rat (acceptor).This group tracheal has epithelial cells.B group,the same way with A group,however,this group has no epithelial cell.C group(control group1),Wistar rat tracheal heterotopic transplantation model with Sodium Chloride,the model subcutaneous implanted in the back of Wistar rat(acceptor).D group(control group2),the same way with C group,however,the tracheal model has no epithelial cell.Toke out the model in different stages after transplant:5 days,10 days,20 days,30 days.Observed the development of OB or not.[Main outcome measures]:Pathological section,trachea obliterate rate.[Results]:A group has obliteration;B group has no obliteration;C and D groups have no obliteration.[Conclusion]:In rat tracheal heterotopic transplantation model,we confirmed that tracheal epithelial cell play important role in development of OB.
