Objective:Observation of rat bone marrow mesenchymal stem cells (BMSCs) transplantation in acute hepatic failure(AHF) in the ability of colonization.Methods:Isolated and cultured in vitro and identification the third generation of BMSCs.BMSCs were labeled with Bromodeoxyuridine(BrdU) AHF animals were induced by D-galactosamine(D-gal).Transplantation of labeled BMSCs(1.5~2.0×10~6),were performed on the AHF rats(group A) /normal rats(group B),another normal group(groupC) were injected with sodium chloride o investigate its gather around the wound site and identify contrbution of bone marrow-derived cells to rat with acute hepatic failture wound contribution in different time(14,21day),Then the mortality of the rats and liver function recovery status were observed. Results:flow cytometry showed that BMSCs expressed CD29,CD44,but no CD34 and CD45 which is not founded in the control groups,brdu labeling efficiency of marrow-derived stem cells was 90%o After cells transplantation there are more BMSCs traced with BrdU were detected in the groop A than B by immunohistochemistry.(P<0.05).Conclusion:BMSCs transplanted through tail vein can successfully reside and homing in normal or in injury liver of rats.But the number of BM SCs homing to liver might be related to the extent of liver injury.
