Objectives: To study the expression of rat insulin-like growth factor-1(IGF-1) in vitro neurons infected with recombinant adeno-associated virus(rAAV) mediated rat IGF-1 gene, and the protection from apoptosis induced by high glucose.Methods: Cloned the rat IGF-1 gene into plasmid pSNAV2.0 applicating molecular cloning technique, and constructed rAAV hybrid serotypes vectors(rAAV2/1-IGF-1) with plasmid pSNAV2.0-IGF-1. When human neuroblastoma cells line(SH-SY5Y) were infected with rAAV2/1-IGF-1, we detected the expression of IGF-1 using RT-PCR and WESTERN BLOT. Furthermore, we studied the protection from apoptosis determined by fluorescence microscope and flow cytometry when SH-SY5Y infected by rAAV2/1-IGF-1 was exposured to high glucose.Results: When SH-SY5Y was infected with rAAV2/1-IGF-1, the expression of rat IGF-1 could be detected by RT-PCR. The results of WESTERN BLOT demonstrated that rAAV2/1-IGF-1 could elevate IGF-1 expression level significantly versus control groups. SH-SY5Y infected by rAAV2/1-IGF-1 could cut down the apoptosis ratio obviously via enhanced IGF-1 expression.Conclusions: Neurons in vitro infected by rAAV2/1-IGF-1 could express IGF-1 significantly, rAAV2/1-IGF-1 could protect neurons from hyperglycemia -induced apoptosis efficiently.
