ObjectTo study this topic in different degrees of damage in osteoarthritis synovial fluid in the joints, IL-1βand TNF-αcontent and cartilage typeⅡcollagen content related to each other. Thus, to further explore the pathogenesis of osteoarthritis mechanism.Materials and Methods1.Laboratory Animals: 20 New Zealand white rabbits, weighing an average of 2.2-2.5kg, male and female are not limited by the Shandong University School of Medicine Center for Animals. Were randomly divided into 4 groups 5 (10 knees).2. Osteoarthritis of the production of animal models: Hulth [1] model is the most commonly used animal model of osteoarthritis.3. IL-1βand TNF-αand collagen typeⅡDetermination: After the success in animal models, 4 weeks, 5 weeks, 6 weeks, respectively, in synovial fluid samples, measuring IL-1βand TNF-αcontent in the articular cartilage andⅡcollagen content.4. Initial processing of specimens: normal, respectively, after the success of animal models and 4,5,6 weeks, the rabbit hindlimb femoral condyle, with 10% paraformaldehyde fixed, 15% neutral EDTA decalcification. Conventional dehydration, transparent, paraffin-embedded into paraffin sections.5.Sample HE dyeing6. Immunohistochemical staining: use of SP on typeⅡcollagen specimens measured.7. Statistical analysis: Data analysis using SAS 8.1 statistical analysis software and variance analysis, statistically significant level of 0.05.Results1. Synovial fluid IL-1βand TNF-αcontent: the control group in rabbit synovial fluid IL-1βand TNF-αcontent is very low, the animal model experimental group 4,5,6 weeks after the measurements were significantly higher than control group .2. Synovial fluid IL-1βand TNF-αcontent: the control group in rabbit synovial fluid IL-1βand TNF-αcontent is very low, the animal model experimental group 4,5,6 weeks after the measurements were significantly higher than control group .3. IL-1βand TNF-αtypeⅡcollagen content and the relevance of IL-1βand TNF-αtypeⅡcollagen content and positively related to each other. Application of analysis of variance, different degrees of damage of articular cartilage in osteoarthritis of typeⅡcollagen differences.ConclusionOsteoarthritis synovial fluid IL-1βand TNF-αand collagen typeⅡwas a positive correlation. Articular cartilage with increasing the degree of injury, IL-1βand TNF-αlevels decreased gradually, while the number of typeⅡcollagen is gradually reduced. That IL-1βand TNF-αon the incidence of osteoarthritis play a role, may be the one of pathogenesis of osteoarthritis.
