ObjectiveTo observe the effect of glucose of different concentrations and its fluctuation on CD11b, IL-6 and TNF-αexpression in monocytes, and to study whether or not osmotic pressure changes and cell apoptosis participate in the process in which intermittent high glucose affects inflammatory factors expressed by monocytes,. THP-1 cells cultured in vitro. The aim is to study the importance of intermittent high glucose in the mechanism of diabetic marcovascular disease, so as to provide theoretical supports and experimental bases for the treatment of. diabetic marcovascular disease.Method1. Monocytes culture: the THP-1 cell strains were thawed on a superclean bench and then grafted to a culture flask which was put in an incubator with 5% CO2(37℃).the cultured monocytes would be stained by Trypan blue to calculate the survival percentage.2. Grouping: we divided, according to glucose concentration and its fluctuation, into three groups, they are the contrast(5 mmol/L),hypertonic glucose(15 mmol/L), intermittent high glucose (5-15 mmol/L),and osmotic pressure controlled (mannitol 5-15 mmol/L)3. Test of inflammatory factors: we used flow cytometry to test CD11b expression on the surface of monocytes and cell apoptosis respectively, and ELISA to test IL-6 and TNF-α, comparing the differences for statistical significance.Result1. CD11b, IL-6 and TNF-αexpression in hypertonic glucose are much higher than those in the contrast.2. CD11b, IL-6 and TNF-αexpression in intermittent high glucose are much higher than those in hypertonic glucose.3. CD11b, IL-6 and TNF-αexpression in osmotic pressure controlled are between those in hypertonic glucose and intermittent high glucose, but compared with them, it is statistically more significant(P<0.05).4. The cell apoptosis are of no statistical significance.Conclusion.1. Glucose of higher concentration enhances THP-1 cells to express more CD11b IL-6 and TNF-α.2. Glucose of intermittent high glucose enhances THP-1 cells express more CD11b IL-6 and TNF-αthan glucose of high concentration.3. The concentration fluctuation of glucose in promoting monocytes to express inflammatory factors is related with osmotic pressure change, but unassociated with cell apoptosis.
