Purposes1.Based on the classical two-cuff technique for rat liver transplantation model,with its own operation continuously be improved to get better mastery of microsurgical techniques.2.In this model,to further explore the basis of natural medicine artemisinin anti-rat liver transplantation the role of immune rejection,efforts to find resistance to artemisinin act on the best dose of immune rejection by the immune rejection against the deeper understanding of to be used for future clinical treatment of artemisinin resistance after liver transplantation immune rejection better basis.Method Two-cuff technique for SD-Wistar rat model of liver transplantation in experimental animals:healthy male SD rats as donors,weight 220~250 g,male Wistar rats as the receptor,weight 250~280 g,for body weight slightly less than the receptor 20~30 g,rat liver transplantation model,and then depending on the time after 5 rats,respectively after 3 days,6 days,9 days of treatment,sampling for aspartate aminotransferase(AST),alanine aminotransferase(ALT),total bilirubin (TBIL),interleukin -2(IL-2),interleukin -4(IL-4) and tumor necrosis factor(TNF-α), At the same time,HE staining of liver tissue evaluation of degree of immune rejection.Results 1.Rat liver transplantation in 100 trials,80 cases of patients survived more than 3 days after surgery,surgical success rate was 80%,the basic agreement with the experimental requirements,not more than three days after surviving the major cause of death was bleeding in rats,hepatic inferior vena cava suture Louxue,thrombosis, and portal vein thrombosis and a few too small and there is variation in vascular death.2.Drug treatment,the groups were significantly different compared to the overall(P<0.01),the control group with high and low doses of artemisinin after AST,ALT, TBIL with time were significantly higher in the blank group high and low dose group were significantly different compared to artemisinin(P<0.01).IL-2,IL-4,TNF-αcompared the two groups was statistically significant difference(P<0.01).High and low artemisinin IL-4 was increased with time,while the control group decreased significantly compared to the two groups(P<0.01).Cyclosporin A group and cyclosporine A + artemisinin group of IL-2,TNF-αand AST,ALT,TBIL with decreased time,while the IL-4 was significantly higher than the two groups,two groups There was no significant difference(P>0.05).Control group and the artemisinin group and the cyclosporine A group and CsA + artemisinin significantly different compared(P<0.01).Cyclosporin A group and cyclosporine A + artemisinin group and the high and low doses of artemisinin were significantly different than(P<0.01).High and low artemisinin between the two groups was not obvious.HE staining and the level of the control group were significantly heavier than artemisinin dose cyclosporine A group and Cyclosporine A + artemisinin group.Control group control group were more severe in high and low artemisinin group,artemisinin group was lower than the level of cyclosporine A and cyclosporine A + artemisinin group,high and low artemisinin between the two groups was not obvious.Conelusionl.Two-cuff technique for accurate and reliable rat model of liver transplantation,liver transplantation can be used as an effective means of experimental studies,and SD(?)-wistar as the ideal model of immune rejection.2.Artemisinin can inhibit IL-2,TNF-αsecretion,significantly reduced the degree of rejection.3.Artemisinin and Cyclosporin A has obvious synergy with this experiment to reduce the dose of cyclosporine A increased the dose of artemisinin,can still obtain satisfactory results.Reasonable match between the two doses of cyclosporin A with the total effect of immune rejection against very specific vision needs further study with dose.4.Artemisinin is effective in promoting the expression of IL-4,and inhibited the immune suppression.5.Medium dose and high dose application of artemisinin can inhibit the immune rejection response in the occurrence of the role of artemisinin in the immune rejection of the maximum dose to be studied further.6.Artemisinin could mechanism of resistance to immune rejection:inhibition of Th1 differentiation may be value added,value-added for Th2 differentiation induced humoral and cellular immunity occurs,thereby reducing the immune rejection response.
