Objective:To investigate the relationship of MTHFRC677T polymorphism, the levels of homocysteine,folate and VitB12 with FGR.Material and Methods:1.sampleIn our study,71 cases of FGR and 60 cases of normal pregnancy,matched for age,height weight,BMI,and gestational week,were recruited. Two groups of pregnant women had no previous hypertensive disease, heart disease, kidney disease, diabetes,liver disease or immune disease, no history of blood transfusion,no history of drug abuse.2. Methods①The mutation of the 677C→T transition of MTHFR was invetigated by polymerase chainreaction-restriction fragment length polymorphism ( PCR-RFLP ) method. Digestion products with 2% agarose gel electrophoresis imaging system, based on restriction map to determine genotypes, analysis of MTHFR gene 677 polymorphism and allele frequencies.②Blood samples were sent to our laboratory,hcy concentration was determined by fluorescence polarization immunoassay(FPIA).And serum folate and vitamin B12 levels were determined with microparticle enzyme immunoassay(MEIA).③All data were analysed with spss10.0 software. Differences in the frequencies of alleles and genotypes were analyzed by Pearson'sχ2 test. Differences of measurement data between two groups were compared by t test,and all data were indicated as x±s ,while homogeneity of variance,using one-way ANOVA, values were considered as significant at p< 0. 05.Results:1. There was no significant difference between the two groups in terms of age,height,weight,BMI and gestational weeks .(P >0.05)2.The frequency of MTHFR C677T T/T(26.8%) was significantly higher than control group(8.3%),The total mutation allele T frequency(42.3%) was obviously higher than control group(19.2%)( P < 0. 05) .3.In FGR group, the concentration of Hcy in MTHFR T/T was obviously higher than MTHFR C/C and C/T, but the concentration of folate was generally decreased ( P < 0.05). Among the three genotypes, there was no significant difference in level of serum VitB12 in FGR group or in level of Hcy, folate and VitB12 in controls.( P > 0. 05).4.The mean level of plasma Hcy in the FGR group ( 13.9±5.4μmol/L) was higher than that in controls(7.3±2.3μmol/L)(P<0.05).5.The mean level of serum folate in the FGR group (12.9±5.2nmol/L) was lower than that in controls( 15.0±5.4 nmol/L)(P<0.05).There was no significant difference in the mean level of vitaminB12 between the two groups(P>0.05).6. According to genotype,we comprehensive analysis of all the data of the two groups, the concentration of Hcy in MTHFR T/T was obviously higher than MTHFR C/C and C/T, but the concentration of folate was lower than MTHFR C/C and C/T ( P < 0.05), Among the three genotypes, there was no significant difference in level of VitB12( P > 0. 05).Conclusions:1. The frequency of MTHFR C677T T/T was significantly higher than control group, the total mutation allele T frequency was obviously higher than control group,the MTHFR C677T polymorphism is associated with of FGR.2. The mean level of plasma Hcy in the FGR group was higher than that in control group,the result showed that elevated homocysteine level is a risk factor of FGR.3. The mean level of serum folate in the FGR group was lower than that in control group ,the result showed that folate is effective in lowing hcy levels in patients with TT and CT genotypes for MTHFR C677T mutation.
