ObjectTo search specific markers for diagnosis and to do some preliminary study about such protein by analyzing the serum proteome of the patients suffering from chronic hepatitis B with different degree of severity.MethodsThe serum of the patients suffering from chronic hepatitis B with different degree of severity was collected and proteome was separated by two-dimensional gel electrophoresis.Then specific proteins were identified by peptide mass fingerprint based on matrix-assisted laser desorption-ionization time of flight mass spectrometry (MALDI-TOF-MS).Within the four proteins identified,we prepared the polyclonal antibody of chain A and using the western blot of serum to verify the result of two-dimensional gel electrophoresis.We also did the immunohistochemical staining with the tissue microarray technology to detect the different expression of this protein in patients with different degree of severity.At last,we cultured 6 cell lines and extracted the total proteins to do the western blot to verify the result of immunohistochemical experiment.Results1.The specific protein chosen from comparing the 2-D page images were identified by mass spectrometry.The proteins with significant difference on expression were Chain A,Interactions Causing The Kinetic Trap In Serpin Protein Folding,HP protein,albumin,isoform CRA_t,and an unnamed protein species.2.Among the serum of the patients suffering from chronic hepatitis B with different degree of severity,the expression of Chain A,Interactions Causing The Kinetic Trap In Serpin Protein Folding was increased along with the severity was worse especially in the patients with hepatic carcinoma.3.The result of the immunohistochemical staining with 6 cases of carcinoma and corresponding adjacent tissues showed that Chain A,Interactions Causing The Kinetic Trap In Serpin Protein Folding almost did not express in the hepatic carcinoma cells.4.The result of the western blot of total protein from a variety of cell lines showed that only Chang cell which was considered as normal control expressed Chain A, Interactions Causing The Kinetic Trap In Serpin Protein Folding.ConclusionUsing progressed two-dimensional gel electrophoresis could be obtained to separate and identify the proteome in serum.In this study,we found 4 different expressed proteins within the patients suffering from chronic hepatitis B with different degree of severity and then we prepared the polyclonal antibody of Chain A,Interactions Causing the Kinetic Trap in Serpin Protein Folding to do more analyses.According to the results, Chain A,Interactions Causing the Kinetic Trap in Serpin Protein Folding almost did not express in the tissues of patients with hepatic carcinoma but the expression in the serum was increased.It might suggest that there are important functions of this protein in patients with hepatic carcinoma and it might be valuable for further explorations.
