UV-B radiation can significantly enhance the contents of anti-toxins (flavonoids, lignans, etc.) in plants, and can also help with the biosynthesis of new bioactive compounds. There are many applications of UV-B radiation in researching the antitoxin of crops, however, few has been reported on the subject of UV-B radiation improving the activity of Chinese medicine ingredients. In order to provide new technical methods of using medicinal resources rationally and improving the quality of medicine, the research revealed the molecular mechanism of UV-B radiation in influencing biosynthesis of the active ingredients at protein level.By using the technology of HPLC-DAD and 2D-Electrophoresis, the research successfully screened out the suitable condition for the UV-B radiation of mulberry leaf, isolated and identified the inducing compounds, and analyzed the differences proteome after radiation. Specific contents include as follows:1. Five significantly changed chromatographic peaks were detected in the extracts of mulberry leaf after induction using HPLC-DAD method. By single factor study, suitable condition for mulberry induction was screened out based on the index of five chromatographic peaks;2. Under optimum condition, five chromatographic peaks were directionally separated, of which two compounds were isolated and their structures were identified;3. The proteomes of mulberry leaves before and after induction were analyzed by 2D-Electrophoresis method, 61 different points were obtained through the processing of Progenesis SameSpots, and eventually 26 differential proteins were identified by MALDI-TOF-MS.The results showed that UV-B radiation technology is a breakthrough of traditional Chinese medicine in biotechnology research. Through this study, authentic data support and methods support can be provided for further revealing the material basis for efficacy and the induction mechanism of mulberry, which is a genuine traditional medicine from Jiangsu and Zhejiang areas.
