Objective To clone mouse IFN-λ2 gene, construct its recombinant adenovirus vector, and to study its biological activity.Methods The expression of mIFN-λ2 in mouse spleen cells was induced by vesicular stomatovirus,mIFN-λ2 cDNA was amplified by RT-PCR and then was subcloned to the vector of pShuttle-CMV. Generation of recombinants by cotransforming the PmeⅠ-cut shuttle plasmids with pAdEasy-1 backbone vectors in BJ5183, Adenovirus was multiplied in 293A cells and the expression of mIFN-λ2 in LA795 cells was identified by immunofluorescence and Western blot.The inhibitory effect of mIFN-λ2 to mouse lung adenocarcinoma LA795 cells was detected by MTT assay and cell survival analysis.Results The sequence of mIFN-λ2 gene was identical with that in GeneBank,the recombinant adenovirus vector was successfully constructed, The inhibitory effect of Ad-mIFN-λ2 and Ad-Lacz to mouse lung adenocarcinoma LA795 cells was detected by MTT assay and cell survival analysis. The conclusion is:the inhibition ration of mIFN-λ2 are to a certain extentConclusions The adenovirus vector of mIFN-λ2 was successfully con-structed and adenovirus recombinants had biological activity.
