[Objective] To evaluate the effects of sandblasting surface, microarc oxidation surface, SBF surface and alkali heat surface on the morphology, proliferation and cell cytoskeleton.[Materials and Methods] The diameter of 15mm, thickness 1mm of pure titanium divided into 12 groups:G surfaces, SB surfaces, AH1-4 surfaces, sbf1-4 surfaces, GMAO surfaces and SBMAO surfaces. Using scanning electron microscopy (SEM), X-ray diffraction (XRD), energy spectrum analysis testing physical and chemical properties of titanium surface. Osteoblast cells were cultured in each group and evaluated by SEM, LSCM, MTT assay, rat osteocalcin (BGP) ELISA assay to compare the group of titanium surface on morphology, proliferation and cytoskeleton.[Results] 1.on the basis of grinding and sandblasting, AH surfaces appeared three-dimensional nano-network structures, SBF surfaces deposited hydroxyapatite granules, MAO surfaces appeared crater shape holes.2.the improved method of primary culture osteoblasts is practicable, efficient and stability.3.Osteoblasts proliferation that SBMAO surfaces is the best one in each group. And BGP detection shows that on day14, MAO surfaces is better than others, andSBMAO surfaces is better than GMAO surfaces.4.Osteoblasts were cultured after 12 hours,LSCM shows that Actin fibers in each group is clearly visible, parallel fibers and cells in the same axis. G surfaces Actin fibers arranged along the scratch direction.SB surfaces osteoblasts coverage in the hole and Actin fibers arranged along the hole edge.SBFl-4 surfaces Actin fibres arranged according to the attachment points, and surrounding the surface of hydroxyapatite.AH1-4 surfaces Actin fibers along the surface of a regular arrangement,and some actin filaments reached to holes.MAO surfaces the actin fibers formed bundles and reached to holes, Ring-shaped arrangement of actin fibers decreased.[Conclusion] 1.the improved method of primary culture osteoblasts is practicable, efficient and effective.2. Different surface roughness on the titanium surface caused by sandblast have certain impact on alkali heat, microarc oxidation and sbf.3.MA0 has the obvious characteristics and it's good for proliferation.4. Observed by LSCM, osteoblast spreading of actin fibers has relation to the surface topography.
