Variation Of Serum Levels Of Interleukin-12p70, Interferon-γ, Tumor Necrosis Factor-α, Interleukin-6 And 10 And It's Clinical Significances In Patients With Dengue Fever

Objective:To investigate the variation of serum level of Interleukin-12p70 (IL-12p70),Interferon-γ(IFN-γ),Tumor necrosis factor-α(TNF-α),Interleukin-6 (IL-6) and Interleukin-10(IL-10) and it's clinical significances in patients with dengue fever(DF).Methods:A total of 40 DF patients and 18 normal controls were enrolled.The serum levels of IL-12p70,IFN-γ,TNF-α,IL-6 and IL-10 were measured with ELISA method.The results were compared between DF patients and normal controls,and various groups based on the course and severity of the disease as well.The relationships among the variations of these cytokines were analysed.The roles of the cytokines on pathogenesis of DV infection and disease development were explored.Results:1.The serum levels of IL-12p70,IFN-γ,TNF-αand IL-10 in DF patients were significantly higher than those observed in the health controls(p<0.001,0.05,0.01 and 0.01;respectively).The serum IL-6 level in DF patients was not significantly higher than those observed in the health controls(p>0.05).2.The serum levels of IL-12p70,IFN-γand TNF-αin incipient DF patients were significantly higher than those observed in convalescence DF patients,all with P value less than 0.01.The serum levels of IL-6 and IL-10 in incipient DF patients were lower than those observed in convalescence DF patients,but the differences were not significant statistically,all with P value more than 0.05.The ratios of serum levels of IFN-γ/IL-6,IFN-γ/IL-10,TNF-α/IL-6 and TNF-α/IL-10 in incipient DF patients were significantly higher than those observed in DF convalescence patients(p<0.001,0.001,0.01 and 0.01;respectively).The serum levels of IFN-γ/IL-6,IFN-γ/IL-10,TNF-α/IL-6 and TNF-α/IL-10 in typical incipient DF patients were significantly higher than those observed in typical convalescence DF patients(p<0.01,0.05,0.05 and 0.01;respectively).3.The serum levels of IL-12p70,IFN-γand TNF-αin severe DF patients were significantly higher than those observed in typical DF patients(p<0.001,0.01 and 0.01;respectively).The serum levels of IL-6 and IL-10 in severe DF patients were lower than those observed in typical DF patients,but the difference were not significant statistically(p>0.05 and 0.05;respectively).The ratios of serum levels of IFN-γ/IL-6,IFN-γ/IL-10,TNF-α/IL-6 and TNF-α/IL-10 in severe DF patients were significantly higher than those observed in typical DF patients(p<0.01, 0.001,0.01 and 0.05;respectively).4.The serum IL-12p70 levels were positively correlated to the serum TNF-αand IFN-γin DF patients(R=0.65,0.72;p<0.001).The serum TNF-αlevel was positively correlated to the serum IFN-γin DF patients(R=0.37,p<0.05).The serum IL-6 level was positively correlated to the serum IL-10 in DF patients (R=0.32,p<0.05).The serum IL-12p70 levels were positively correlated to the serum TNF-αand IFN-γin acute stage of dengue fever patients(R=0.56,0.59; p<0.05).The serum IL-12p70 levels were positively correlated to the serum TNF-αand IFN-γin convalescence stage of dengue fever patients(R=0.55,0.77; p<0.01,0.001).The serum IL-12p70 level was positively correlated to the serum IFN-γin severe dengue fever patients(R=0.60,p<0.05).The serum IL-12p70 level was positively correlated to the serum TNF-αin typical dengue fever patients(R=0.69,p<0.001).Conclusion:DV infection may results in the variation of serum IL-12p70,IFN-γ, TNF-α,IL-6 and IL-10 levels in DV-infected patients.Thees cytokines may be involved in the pathogenesis of dengue fever.The imbalance of Th1/Th2 type cytokines in the process of DV infection may plays an important role.Serum IL-12p70 in patients with dengue fever,may promote the secretion of Th1 type cytokines and inhibit the secretion of Th2 type cytokines,which play a dominant Th1 response.
