This paper contains two researches on Salviae Miltiorrhizae. The first part is to develop a simple and accurate method for simultaneous determination of two lipophilic components and three hydrophilic components of S. Miltiorrhizae. This method was used for determining the five bioactive components in 11 clonal materials and in different parts, diameter and root colour of S. miltiorrhiza.The other part is about the research focusing on the expression of gene SmAACT, SmDXR, SmHMGR and SmCPS, which are related to the biosynthesis pathway of tanshinone compounds, in different tissues through real-time PCR...Main contents as follows:①The chemical composition and key enzyme genes of tanshinone biosynthesis pathway of Salviae S. Miltiorrhizae were reviewed;②To develope a new simple and accurate method for simultaneous determination of three hydrophilic components and two lipophilic components of S. Miltiorrhizae. The developed method is in good repeatability, precision and stability;③The quality research on S. Miltiorrhizae of different origin;④The content of these five compounds in different parts, diameter and root color of S. Miltiorrhizae were analyzed.;⑤Gene SmAACT,SmHMGR, SmDXR can express in all the ten chosen tissues, while gene SmCPS cannot express in petal, cortexs and sepal.The results show that:The content of hydrophilic components and lipophilic components in clonal materials of S. Miltiorrhizae are quite different, the DC003, DC004 and DC007 samples of three regions are high of lipophilic components; The DC007 samples have the highest hydrophilic components. The data analysis showed that Danshensu, salviamolic acid B and tanshinoneⅡA levels of clonal materials were significant differences. The DC003, DC004, DC007 can be used as raw materials for hybridization to filter out new S. Miltiorrhizae of higher hydrophilic components and lipophilic components; The contents of Danshensu, salvianolic acid B, cryptotanshinone and tanshinoneⅡA in the cortexs were higher than in the wood; The thinner the root was, the higher its contents of cryptotanshinone and tanshinone IⅡA were, and the lower its content of salvianolic acid B was. Protocatechuic aldehyde was concentrated in the head of the root. When the color of root powder was closer to red, and the deeper the color, its contents of hydrophilic components and lipophilic components were higher. The exrpression results of gene SmAACT.SmHMGR, SmDXR and SmCPS suggest that SmCPS may play a regulatory role in the biosynthetic pathway of tanshinone compound.This paper develop a new method for simultaneously determine three hydrophilic and two lipophilic bioactive components in S. Miltiorrhizae. It provided a quick method of multiple bioactive components simultaneous determination for S. Miltiorrhizae quality analysis. To compare the quality and the chemical components of different parts, diameters and root colours of S. miltiorrhiza. Bge.from different areas,for the breeding of new varieties of S. Miltiorrhizae, GAP normalization cultivation and use of various drug companies to provide the theoretical basis for the original medicinal of S. Miltiorrhizae. In this paper, research of four key gene expression patterns of tanshinone biosynthetic pathway, help people more clearly know that the level of transcription of molecular mechanism of tanshinone biosynthetic pathway, for the regulation of biosynthesis of tanshinone provide theoretical basis.
