Objective:To study the killing effect of different temperatures and concentrations of cisplatin thermo-chemotherapy on the human hepatic carcinoma cells hepg2 in the blood and the influence on the biological activity of red blood cells.Methods:Add the cultured hepatoma cells hepg2 to the concentrated red blood cells,then add DDP and heat treatment.We set up three DDP working concentrations (respectively 50μg/ml,100μg/ml,200μg/ml),heat treatment time is located at 37℃and 42℃,the heating time are 30 min.the tumor cells are separated by density gradient centrifugation from blood.Assay the inhibitory rate of tumor cells by MTT,check the tumor clone colony to calculate colony-forming rate,determinatin the activity of the erythrocyte Na+-K+-ATP enzyme.Results:Compared with the control group,liver cell lines hepg2 with different temperatures and different concentrations of cisplatin in the number of viable cells after treatment were significantly reduced,and also with a concentration-dependent decrease (P<0.005),heat can inhibit the growth of tumor cells,and also can enhance the killing effect of Platinum on the hepatic carcinoma cells hepg2; the colony formation rate of tumor cell lines in each group were significantly reduced, decreases along with the increasing heat treatment temperature and the increasing concentration of platinum (P<0.005).42℃heat treatment group and the cisplatinum at 200μg/ml when no cell colony formation.After the heat treatment with cisplatin in each group the activity of Na+-K+-ATP enzyme on erythrocyte membrane decreased,but compared with the control group, does not become significantly lower (P>0.05),and also each group don't change significantly in the morphology of RBC.Conclusion:1. Cisplatin combined with heat treatment show signifificant inhibition on the hepato cellular carcinoma cell lines,the heating can increase the sensitivity of cells to cisplatin and enhance cytotoxicity of cisplatin; cisplatin at the concentration of 200μg/ml and the joint action of 42℃heat treatment for 30min,the tumor cells appear more than 99% clearance rate,and no tumor cell colony come into being formation.2. cisplatin and heat treatment did not produce significant effect on morphology of red blood cell and the activity of Na+-K+-ATP enzyme.
