Expression And Significance Of WWOX Gene In Leukemia | | Posted on:2011-06-07 | Degree:Master | Type:Thesis | | Country:China | Candidate:Z L Chen | Full Text:PDF | | GTID:2144360305452419 | Subject:Clinical Laboratory Science | | Abstract/Summary: | PDF Full Text Request | | Objective①To investigate the expression and significance of WWOX tumor suppressor gene in acute leukemia.②To preliminary explore the correlation of WWOX and Bcl-2 protein in acute leukemia.Method①Reverse polymerase chain reaction(RT-PCR) was used to analyze the expression of WWOX mRNA in 72 leukemia patients and 20 control cases.②Immunohistochemistry was used to test the expression of WWOX, Bcl-2 in 72 leukemia patients, 20 control cases, and to preliminary explore the dependability of WWOX and Bcl-2 protein in leukemia.Result①The positive rate of WWOX mRNA in leukemia(52.77%) wasobviously lower than that in control cases(90.00%) (P<0.01); Comparison of the group of the the newly diagnosed cases, relapsed cases and control cases in acte leukemia, we found that the WWOX mRNA positive rate of newly diagnosed cases(39.39%) and relapsed cases(36.36%) in leukemia were very significantly lower than that of control cases (90.0%) (P<0.05);②The positive rate of WWOX mRNA between acute myeloid leukemia and acute lymphocytic leukemia was significantly lower than that of control cases(P<0.05), but there were not significantly different(P >0.05); ③Comparison of the group of the newly diagnosed cases, relapsed cases and remission cases in acute leukemia, we found that the WWOX mRNA positive rate of newly diagnosed cases(39.39%) and relapsed cases(36.36%) in leukemia were very significantly lower than that of remission cases(75.00%) (P<0.05),but there was not significantly difference between newly diagnosed and relapsed cases,between remission cases and control cases (P >0.05);④Analyzing the different influence factors among the groups of newly diagnosed cases , we found that there was not significant difference about the WWOX mRNA positive rate between older(>40) and younger(≤40) patients, between male and female patients, between patients with WBC>50×109/L and WBC≤50×109/L, between patients with higher( >50%) and lower(≤50%) immature cells in the peripheral blood, between patients with or without hepatosplenomegaly , between patients with or without glandula lymphatica (P >0.05);⑤The results of Immunohistochemistry demonstrated that the positive rate of expression of WWOX protein in leukemia patients was significantly lower than that of the control cases(P<0.01), while that of Bcl-2 protein in leukemia patients was significantly higher than that of the control cases(P<0.01); But there was not significantly difference between the leukemia patients in AML and ALL groups in the positive rate of expression of WWOX protein and Bcl-2 protein(P >0.05);⑥Comparison of the groups of the newly diagnosed cases and remission cases and analysis the positive rate of expression of WWOX protein and Bcl-2 protein, we found that there were statistically significant difference (P <0.05);⑦The positive rate of the expression of WWOX protein in acte leukemia patients was positively correlated to the expression of WWOX mRNA (r=0.653,P<0.01), while the positive rate of the expression of WWOX protein in acte leukemia patients was negatively correlated to that of Bcl-2 protein (r=-0.363,P<0.01).Conclusion1,The expression of WWOX in acute leukemia patients was usually absent or reduced, it may take an important effect in the pathogenesis and development of leukemia.2,Compared with the negative expression of WWOX mRNA in newly diagnosed patients with leukemia,the positive expression of WWOX mRNA in newly diagnosed patients with leukemia have a higher remission rate, so we can conclude that WWOX may be regarded as one of the new biological parameters in diagnosis of leukemia and efficacy test.3,The expression of WWOX protein in leukemia patients is negatively correlated with Bcl-2, these two proteins may take effect in the pathogenesis and progress of leukemia. | | Keywords/Search Tags: | Leukemia, WW domain-containing oxidoreductase (WWOX), B-cell CLL/lymphoma 2(Bcl-2) | PDF Full Text Request | Related items |
