Objective The ERAP1 protein was isolated and identified from lymphatic metastasis-associated differentially expressed protein in ovarian carcinoma cell lines with directional highly lymphatic metastasis by the proteome technology. To realize the the role of ERAP1 protein in the ovarian cancer lymph node metast-asis process and analyze its relations to the ovarian cancer patient clinical patho-logy as well as to the ovarian cancer patient diagnosis and the judgment prongo-sis value. The protein expression level of ERAP1 was detected in human ovarian cancer cell lines among non-directional (SKOV3) and directional highly lym-phatic metastasis (SKOV3-PM2, SKOV3-PM3, SKOV3-PM4), the ovarian cancer transplanted tumor primarily focus and the ovarian cancer lymph node metastasis of nude mouse,the human ovarian cancer primarily focus and the ova-rian cancer lymph node metastasis and different ovarian cancer patient blood se-rum. To realize the the role of ERAP1 protein in the ovarian cancer lymph node metastasis process and analyze its relations to the ovarian cancer patient clinical pathology as well as to the ovarian cancer patient diagnosis and the judgment prognosis value.Methods The cytoblastema protein was extracted from ovarian carcinoma cell lines with directional and non-directional highly lymphatic metastasis. The expressions of ERAP1 protein in the human ovarian cancer cell lines between non-directional and directional highly lymphatic metastasis was determined by western-blot. The localization and expreesion of the ERAP1 were performed by immunohistochemistry in the ovarian cancer transplanted tumor primarily focus, the ovarian cancer lymph node metastasis of nude mouses, the 20 cases human ovarian cancer primarily focus and the ovarian cancer lymph node metastasis. The expressions of ERAP1 protein in 40 clinical patients' blood serum was determined by ELISA.Results The expression level of ERAP1 protein is down-regulated in SKOV3, SKOV3-PM2, SKOV3-PM3,SKOV3-PM4 cell sublines,and the level of ERAP1 in SKOV3 cell is higher than the other three cells (P<0.05). The positive reaction cells assume yellow or brown pellet in cytoplasma by imm-unohistochemistry. It assumes the declining trend in the human ovarian cancer cell lines between non-directional and directional highly lymphatic metastas-is,the ovarian cancer transplanted tumor primarily focus of nude mouse and the ovarian cancer lymph node metastasis of nude mouse, the human ovarian cancer primarily focus and the ovarian cancer lymph node metastasis. There are sig- nificant differences between these groups(P<0.05). The level of ERAP1 in patient blood serum of terminal patient from ovarian cancer is lower than the group of benignity and the group of early ovarian cancer patient. The results showed that there was differences significantly between these groups (P<0.05).Conclusions The deficits or down-regulated expression of ERAP1 protein is closely related with ovarian cancer lymph node metastasis.
