Objective To study the influence of Hydroxy Safflor Yellow A on the migration and VEGF expression of retinal and choroid endothelial cell in Rhesus monkey under high glucose condition.Methods The RF/6A cells were bought from Shanghai Bank of Academia Sinica ,we cultured the cells in out-of-body condition and then devided them into group of NG+H0, NG+H18, NG+H37, NG+H73 contained with nomal glucose of 5.5mmol/L and added with HSYA of 0mg/L,18mg/L,37mg/L , 73mg/L and group of HG+H0, HG+H18, HG+H37,HG+H73, contained with high glucose of 22mmol/L and added with HSYA of 0mg/L,18mg/L,37mg/L,73mg/L. We took the pictures at 0h,24h,48h,72h with inverted microscope by cell scratch healing assay to study the migration distances which react the capability of cells migration ; observed the influence of Hydroxy Safflor Yellow A on the VEGF expression of cells under nomal and high glucose condition with half fix quantified reverse transcription-polymerase chain reaction (RT-PCR).Spss13.0 package was used for statistical analysis .Values were given in mean±SD ,statistical significance was evaluated with the one-ANOVA.Difference at p<0.05 were considered to be statistically significant.Results 1, Cell scratch healing assay:a.Compared with HG+H0 the cells of NG+H0 group migrated longer distances at 48h and 72h .b.The results between the NG+H18, NG+H37, NG+H73 and NG+H0 groups were no differences,but HG+H18,HG+H37,HG+H73 group cells migration distanse were longer than NG+H0, and the results were related to the concentration of HSYA. 2, Half fix quantified reverse transcription-polymerase chain reaction: HG+H0 group VEGFmRNA was more than NG+H0 group at 24h, 48h, 72h. b. At 48h, 72h, compared with NG+H0 group, HG+H37 group and HG+H73 group VEGFmRNA expression are fewer ,and the results were related to the concentration.Conclusion HSYA may remarkably inhibit the migration and VEGF expression of RF/6A with high glucose,The mechanism may be related to the effect of HSYA on VEGF expression of RF/6A.
