Objective:To assess the proliferation and apoptosis effect of honokiol on human non-small cell lung cancer A549 cells in Vitro.Methods:MTT assay was carried out to observe the anti-proliferation effect of indicated honokiol (0,20,40,60,80umol/L) on A549 cells in Vitro. The apoptosis of A549 cell induced by honokiol was detected by AnnexinV-FITC/PI double-labeling and low cytometry. The expression of Caspase-3,Caspase-9 was examined by western-blot.Results:1 The inhibitory action of A549 cell growth dealing with honokiol was observed and showed as a dose-dependent manner. The inhibitor index of different concentration group (0,20,40,60,80umol/L) reached to 0%,3.25±4.59%,11.90±2.55%,50.30±3.67%,81.70±7.49% respectively. And the later two groups showed significant difference from the control group.2 The apoptosis index of A549 cell was improved as the dose of honokiol increasing. The apoptosis rate of the cell A549 treated with 0,20,40,60 and 80umol/L honokiol were 0%,8.57%,15.61%,38.40%,59.75%respectively.3 The up-regulation of Caspase-3 and Caspase-9 was detected by western-blot assay. The expression diversity between different dozes was also obvious.Conclusion:Honokiol could inhibit the human non-small cell lung cancer A549 cells proliferation in vitro and induce the apoptosis. The apoptosis effect may relate to the activation of Caspase-3,Caspase-9.
