ObjectiveSince the discovery of the antitumor activity of cisplatin in 1969. Platinum complexes had become one of the most widely utilized antitumor drugs in the world. And oxaliplatin is the first platinum complex that highly effective in treating colorectal cancer and has low level of cross-resistance between cisplatin and carboplatin. But oxaliplatin still has several disadvantages:cause nausea,vomiting, bone marrow suppression, neurotoxicity et al. In this study was conducted to identify the in vitro antitumor activity and the influence on cell cycle and cell apoptosis of two novel Platinum (â…¡) complexes against SW620 cell line, and meanwhile compared these effects with oxaliplatin.MethodsThree antitumor agents was administered to colonic cancer cell line of SW620 at different concentrations for 24,48,72h,and then the cell proliferation inhibition rate was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl (MTT) assay. And the analysis of the cell cycle was performed by flow cytometry. Cell apoptosis was examined by means of annexin V-FITC binding assay. Changes in morphology were observed by microscope and scanning electron microscopy.ResultsOur data indicate that both concentration and duration of exposure are important factors for these drugs cytotoxicity. And efficacy can list as follows:Pyridine> Pyrazine>Oxaliplatin.ConclusionsBased on the results of this study we conclude that the in vitro antitumor activity of the two novel type platinum(â…¡) complexes against SW620 cell line was greater than oxaliplatin. And the targets are block the cell cycle and induces cell apoptosis..
