Aim:Obesity is a complex, multifactorial chronic disease frequently associated with cardiovascular risks, hyperlipidemia, high blood pressure, and the insulin resistance that appears to be central to the pathogenesis of TypeⅡdiabetes. The aim of the present study was to establish an obesity rat model to uncover proteins differentially expressed in the adipose tissue of obese mice in early time.Methods:Fifty 4-weeks male C57BL/6 mice divide into two groups due to be fed either a control low-fat diet (LF; 10% saturated fat) or a high-fat diet (HF,58% saturated fat) for durations ranging between 1-14 wk. TC,TG,HDL-C,LDL-C,fasting plasma glucose, uric acid and fasting insulin will be test and epididymal adipose tissue will be taken at 0wk,4wk,14wk. IPGTT will be test at 4wk,11wk,13wk..Then two-dimensional gel electrophoresis and MALID-TOF-TOF were used to identify the differentially expressed protein of epididymal adipose tissue between obesity and control groups in 4wk and 14wk..Results:The body weight between HF group and LF group make significantly difference at lwk (p<0.05). There is no significantly difference of IPGTT(0min,15min,30min,60min),fasting plasma glucose and fasting insulin between HF group and LF group, but significantly difference of IPGTT(120min) TG, TC, HDL-C, LDL-C and urine acid between HF group and LF group at 4wk. There is no significantly difference of IPGTT(0min,15min,30min,60min) between HF group and LF group, but significantly difference of IPGTT(120min) between HF group and LF group at 11wk. There is no significantly difference of IPGTT(0min,60min) between HF group and LF group, but significantly difference of IPGTT(15min,30min,120min) between HF group and LF group at 13wk.There is significantly difference of fasting plasma glucose, TG, TC, HDL-C, LDL-C and urine acid between HF group and LF group, but no significantly difference of fasting insulin between HF group and LF group at 14wk.Two-dimensional gel electrophoresis display:there is no different point between HF group and LF group at 14wk above 1.5.But there is 18 different points between HF group and LF group at 14wk between 1.2 and 1.5,17 of them is up-regulation, one of them is down regulation. There is 10 different points between HF group and LF group at 4wk above 1.5,9 of them is up-regulation, one of them is down regulation. There is 26 different points between HF group and LF group at 4wk between 1.2 and 1.5,25 of them is up-regulation, one of them is down regulation. Matrixassisted laser desorption ionization-time of flight-mass spectrometry were used to identify proteins differentially expressed in WAT between HF group and LF group in 4 wk and 14 wk .Our studies reveal 16 proteins significant changes in obesity.Conclusion:The results of biochemistry display:the diet induced obesity is successful, which contains the obesity and metabolic syndrome. 4wk as the early phase of obesity which make a significant difference of hyperlipoidemia, abnormal carbohydrate tolerance and urine acid between HF group and LF group. This rat model is a diet induced obesity, which is similar to human obesity. The 14-3-3 proteinγand Pinl significant changed in early phase of obesity may be a new marker or protectin of diet induced obesity.
