Objective:To investigate the effects of different doses erythromycin on mucous secretion in ileal neobladder of rats.Methods:Female SD rats(weight 220-250g) were randomized into 4 group, NS group(control group); dehydrated alcohol group; high doses erythromycin group; low doses erythromycin group. The all rats were undergo ileal neobladder. Drug's (erythromycin) metrology is according to preliminary test; Then filling drug after the operation a week later.Collecting the urine samples in ileal neobladder before and after filling drug respectively. All animals were killed after the last collecting the urine samples. The ileal neobladder tissues were evaluated by HE,electron microscope,and total protein determination in urine by BCA method.Results:1. Effects of histomorphology:There is no obviously injury the mucosa of ileal neobladder in High dose erythromycin group and has no obviously difference compared with control group. There is injury the mucosa of ileal neobladder in dehydrated alcohol group and has obviously difference compared with control group.2. Effects of the Ultrastructure of goblet cells:There is no obviously injury the ultrastructure of beaker cells in High dose erythromycin group. There is no obviously difference between High dose erythromycin and control group.There is injury the the ultrastructure of beaker cells in dehydrated alcohol group and has obviously difference compared with control group. 3. Effects of secretion of goblet cellsl:Detected the protein level of each group using BCA method.The high dose erythromycin group's mucus levels is less than NS control group.But there is no significant difference between Low dose erythromycin group and NS control group.4. Effect of amount of goblet cells:the number of goblet cells has obviously difference between high dose erythromycin group and NS control group, high-dose erythromycin group significantly reduced the number of goblet cells. But Low dose erythromycin group has no difference with NS control group.Conclusions:1. Our experiments successful establish the model of rat ileal neobladder and can complete target detection of our required.2. Erythromycin inhibit goblet cell secreting mucus in ileal neobladder and dose no damage structure of goblet cells, moreover it can reduced the amount of beaker cells of ileal neobladder. the specific mechanism needs further study.
