Isodon rubescens is one of the most widely used folk medicines in China, its scientific name is Isodon rubescens (Hemsl) Hara, Health department approvded its dried leaves and above ground as Chinese herbal medicine since Isodon rubescens taste bitter,it can clear away the heat-evil and expell toxin. Its leaves have been used for treatment of esophageal cancer for long time. The phytochemical and pharmacological studies have domenstrated that diterpenoids oridonin is the major active constituent from Isodon rubescens and have been discovered to be responsible for its antiangiogenic activity. The main consituent is kaurene diterpene and there some triterpene and volatile oil left. Good role of Isodon rubescens and its main active ingredient attrated extensive attention from scholars. Many scholars have carried out studies on this nearly 30 years.In this paper, we mainly designed the acute toxicity, chronic liver injury, long term toxicity. This confirmed that it is safe to take Isodon rubescens extractive orally and it has little toxicity long term and protection for injury of liver.we also designed injury liver by alcohol and the paper is composed of five components.The first part is the preface which summarizes the research of liver function injury determination index and progress research in the effect of Chinese herbal medicine on liver oxidative damage.The second part is the IRE tolerance test, from the test we gain the maximum tolerance dose is 40g/kg/d, which provide basis for further experiments.The third part is the IRE acte toxicity test. Acute liver injury of mice was induced by intraperitoneally (i.p.) injection of CCl4; we treat the mice with IRE high, middle and low dosage successive administration for 15 days. The activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum, maleic dialdehyde (MDA) concentration and the activities of superoxide dismutase (SOD) were detected. It can be concluded that IRE markdely reduced the serum ALT and AST activites, reduced the content of MDA and increased significantly the activities of SOD in serum(P<0.01 or P<0.05).The consequence prompt IRE can resist oxidative damage and free radical.The fouth part is the IRE chronic toxicity test. Chronic liver injury of mice was induced by intraperitoneally (i.p.) injection of CCl4, IRE small-, middle-, large-dose groups were administered with IRE at different doses for 8 weeks.The results proved that IRE can degrade significantly the liver weighty increase induced by CCl4 and the level of serum ALT, AST, TP and ALB, reduced the content of MDA and increased significantly the activities of SOD in serum. There has no significant difference between IRE and CLBRT. We conclude that IRE has protective effects on the expermental chronic liver injury induced by CCl4 in mice.The fifth part is the IRE long term toxicity. Rats of three experimental group were given IRE (i.g.) at doses of 40, 20, 10 g/kg for 12 weeks, hematological indexes, biochemical indexes for blood and pathological changes of rats were obseved. The heart, the liver, spleen, lung, kidney were observde with naked eyes and under microscope. No accumulative toxicity was observed in the expermental study on long time toxicity of IRE. We draw a conclusion that it is safe to take Isodon rubescens extractive orally and it has little toxicity.
