Objective:To study the effects of Edaravone on the protein and mRNA expression of brain derived neurotrophic factor (BDNF) and nerve growth factor (NGF) in the permanent focal cerebral ischemia tissues of rats.Methods:Healthy male Wister rats were taken for this study project. According to the procedure of Zea-Longa, the rat model with permanent cerebral ischemia was established by rat middle cerebral artery obstructed (MCAO) with a nylon thread. Operated rats were randomly divided into model control and edaravone treated groups (n=20 for each). By intraperitoneal injection, model control group rats were given physiological saline 3mg/kg once daily for 3days, and also edaravone treated group rats were given edaravone 3mg/kg once daily for 3days.5 rats in each group were taken to tetrazolium chloride (TTC) staining for macroscopic observation of cerebral infarction area, the rest samples were processed by immunohistochemistry to evaluate effects of edaravone on BDNF and NGF expression, hybridization in situ to evaluate effects of edaravone on BDNF mRNA and NGF mRNA expression. SPSS 12.0 for statistical analysis, it was P<0.05 as having statistical significance.Results:1. Grade of neuropathological function Pre-treatment, comparing edaravone treated group and model control group, there did not have significantly neuropathological function difference (P>0.05).3 days later, compare with model control group, the rats of edaravone treated group gain significant improvement of neurological deficit (P<0.05).2. The expressions in brain of BDNF positive and BDNF mRNA positive Compare with model control group, in edaravone group, there was a significant expression up-regulation to BDNF and BDNF mRNA in the peripheral around infarction and cornu ammonis or hippocampus area (P<0.05). However in the infarction area, the expressions of BDNF and BDNF mRNA had no significantly statistical difference (P>0.05).3. The expressions in brain of NGF positive and NGF mRNA positive Compare with model control group, in edaravone group, there was a significant expression up-regulation to NGF and NGF mRNA in the peripheral around infarction and cornu ammonis or hippocampus area (P<0.05). However in the infarction area. the expressions of NGF and NGF mRNA had no significantly statistical difference (P>0.05). Conclusion:1. Edaravone can significantly improve the neurological function of rats after ischemia.2. Comparing to control group, edaravone can significantly up-regulate the expressions of BDNF and NGF in the peripheral around infarction and cornu ammonis or hippocampus area.3. Edaravone maybe protect from the ischemia injury through this mechanism. And so on, it provides more extensive experimental evidence for the clinical application of the medicine.
