Objective:To study the GSK-3βand CD44v6 expression in human colorectal tumors and its significance in tumors development process, to explore the GSK-3βand CD44v6 relevance.Methods:Paraffin-embedded specimens were collected in 2009 from the department of pathology, the first affiliated hospital of Shanxi Medical University. The specimens included 20 cases of normal colorectal mucosa,19 cases of colorectal adenomas and 45 cases of CRC. We detected the expression of GSK-3βand CD44v6 protein in the three groups by immunohistochemristry (Envision method) and analyzed the relationship with the clinicopathological parameters and the relevance of GSK-3βand CD44v6 in CRC.Results:①he GSK-3βin the CRC and colorectal adenoma and normal intestinal mucosa were expressed. In the normal intestinal mucosa, the positive expression of GSK-3βwas found in 1 of 20(5%) cases. In the colorectal adenoma, the positive expression of GSK-3βwas found in 8 of 19(42.1%) cases. In the CRC, the positive expression of GSK-3βwas found in 35 of 45 (77.8%) cases, the expression of GSK-3βin CRC was significantly higher than it in the colorectal adenoma and normal intestinal mucosa, there was significant differences among the three groups(P<0.05). In CRC group, when the tumor size was≤3cm, the frequency of the positive expression of GSK-3βwas 100%, when the tumor size was>3cm, the rate was 72.9%. In the well-differentiated glandular cancer, the frequency of the positive expression of GSK-3βwas 100%, In the moderately differentiated glandular cancer, the rate was 75%. In the poorly differentiated glandular cancer, the rate was 71.4%. In cases with the lymphoid node metastasis, the frequency of the positive expression of GSK-3βwas 77.3%. In cases without the lymphoid node metastasis, the rate was 78.3%. In the stage A+B, the frequency of the positive expression of GSK-3p was 78.3%. In the stage C+D, the rate was 77.3%. The expression of the GSK-3βin CRC was not correlated with Dukes stage, tumor size, lymph node metastasis and histologic differentiation(P>0.05). In the colorectal adenoma group, the frequency of the positive expression of GSK-3βwas 33.3%,46.2%and 33.3%in the mild atypical hyperplasia, moderate atypical hyperplasia, and severe atypical hyperplasia, respectively. There were no significant differences in expression of there groups (P>0.05).②In the CRC, the positive expression of CD44v6 was found in 25 of 45 (55.6%) cases. In the colorectal adenoma and normal intestinal mucosa, the CD44v6 was not found. The expression of CD44v6 in CRC was significantly higher than that in the colorectal adenoma and normal intestinal mucosa(P<0.05). In CRC group, tumor size was≤3cm, the frequency of the positive expression of CD44v6 was 50%, and tumor size was> 3cm, the rate was 56.8%. In the well-differentiated glandular cancer, the frequency of the positive expression of CD44v6 was 57.1%. In the moderately differentiated glandular cancer, the rate was 41.7%. In the poorly differentiated glandular cancer, the rate was 78.6%. In cases with the lymphoid node metastasis, the frequency of the positive expression of CD44v6 was 73.9%. In cases without the lymphoid node metastasis, the rate was 44.5%. In the A+B, The frequency of the positive expression of CD44v6 was 44.5%. In the C+D, the rate was 73.9%.The expression of the CD44v6 in CRC was not correlated with tumor size and histologic differentiation(P>0.05), it was correlated with lymph node metastasis(P<0.05).③he positive expression of GSK-3βand CD44v6 was found in 21 cases. The negative expression of them was 5 cases. The positive expression of GSK-3βand the negative expression of CD44v6 were 14 cases. In CRC, the expression of GSK-3βand CD44v6 were uncorrelated (P >0.05).Conclusion:①The GSK-3βwas involved to settle the occurrence and development of colorectal tumor.②The CD44v6 was involved to progression of colorectal tumor, and it correlated with lymph node metastasis.③In CRC, the expression of GSK-3βand CD44v6 were uncorrelated (P >0.05).
