[Objective] To explore the expression of HMGB1 and RAGE protein in esophageal squamous cell carcinoma(ESCC) and their functions in the prognosis of ESCC.[Methods] We measured HMGB1 and RAGE protein expression in 80 specimens of esophageal carcinoma and 40 specimens of normol esophageal tissue with immunohistochemistry,and analyzed the relationship between these protein expression and the clinicopathological parameters ,and explored the prognosis in ESCC. 46 specimens of esophageal carcinoma and 46 specimens of normol esophageal tissue as well as Western blot analysis to detect the expression of HMGB1 and RAGE.[Results]1. The positive rates of expression of HMGB1 and RAGE in specimens of esophageal carcinoma and normol esophageal tissue were 47.5%vs17.5%,72.5%vs42.5%, the difference had statistical significance.The expression of HMGB1 and RAGE were positively correlated with infiltration degree,lymphatic metastasis and TNM stage, negatively correlated with the degree of cancer differentiation(p<0.05); the expression of HMGB1 positively correlated with that of RAGE(r=0.35,P=0.001).2. Through The Western blot analysis we relief the positive rates of expression of HMGB1 and RAGE in specimens of normol esophageal tissue,esophageal carcinoma without lymphatic metastasis and esophageal carcinoma with lymphatic metastasis had a growing trend(P=0.000); With the degree of cancer differentiation turn down,the positive rates of expression of HMGB1 and RAGE also had a growing trend(P=0.000). 3. The positive expression of HMGB1 and RAGE in ESCC had poor prognosis,COX regression indicated HMGB1 was an independent prognostic marker of ESCC.[Conclusion] HMGB1 and RAGE expression have close relations with the clinical pathophysiological characteristics in ESCC,Maybe the protein correlated with the local invasion and distant metastasis of ESCC. HMGB1 expression may be used as an important survival predictor in ESCC.
