Objectives To investigate the distribution of interleukin-16 polymorphism in Guangxi healthy population and patients with nasopharyngeal carcinoma, analyze the association of IL-16 SNPs rs11556218T/G,rs4778889T/C,rs4072111C/T with NPC.Methods Case-control study of 75 patients with NPC and 75 healthy population in guangxi,the serum level of IL-16 was measured by double antibody sandwich method of ELISA,and the genotype and allele of IL-16 SNPs rs11556218T/G,rs4778889T/C,rs4072111C/T were identified by PCR-RFLP and Sequencing; the haplotype of IL-16 polymorphism was performed using PHASE software; the Hardy-Weinberg equilibrium, genotype, allele and haplotype frequencies were compared between NPC group and control group using the SPSS 13.0 software.Odds ratio(OR) and 95% confidence intervals(CIs) were calculated to assess the relative risk.Results 1.The serum IL-16 level in NPC(40.22±9.09pg/mL) was higher than that of the controls(28.31±5.28pg/mL)(P=0.00).2.The genotype distribution of IL-16 polymorphism was compatible with the Hardy-Weinberg equilibrium in NPC group and control group(P>0.05).3. The rs11556218 polymorphism was found in both caes and controls. There was a significant difference of TG genotype between NPC group and control group, but no significant difference was found in distribution of GG genotype, so the TG genotype was associated with a significantly higher risk of NPC as compared with the TT genotype(OR=2.05; 95%CI, 1.04-4.01 P=0.037). Using the T allele as a reference, a significant correlation was found between the presence of G allele and a risk of developing NPC(OR=1.79; 95%CI, 1.06-3.61; P=0.027). The polymorphisms of rs4778889 and rs4072111 were found in both NPC group and control group, but genotype and allele frequencies ot these two sites in NPC patients were not significantly different from those in control group(P>0.05).4.The distribution of the three SNPs in Guangxi population is not significantly different in Asian subjects, but is significantly different in European n and Nigerian population.5. The presence of G genotype of rs11556218 was associated with a significantly higher serum level of IL-16.6. GTT and GCC haplotypes in the NPC group were higher than that in the control group, there were significant differences of these two haplotypes distribution between two groups(P=0.020; P=0.024).Conclusion The rs11556218T/G, rs4778889T/C and rs4072111C/T polymorphisms of IL-16 were found in guangxi population.The rs11556218T/G polymorphism was significantly associated with the susceptibility to NPC.The TG genotype and G allele were associated with a significantly higher risk for developing NPC(OR=2.05; OR=1.79). GTT and GCC haplotypes were more susceptible to NPC. |
