Zizyphi spinosi semen (ZSS) is the seed of Ziziphus jujube Mill. var. spinosa (Bunge) Hu ex H.F. Chou (Rhamnaceae), which distributes to the heart,liver and hall bladder channels.The total flavonoids of the ZSS extract are the main pharmacological active compounds. Therefore, It is potential significance for further development of studing systematicly on the chemical composition of ZSS. And it provides a new method of isolating and purifying for the further development of ZSS.Therefore, it builds up foundation for development.Firstly,we research the process of extraction of flavonoids from ZSS, and compare the content of total flavonoids extracted by reflux extraction and ultrasonic extraction, determined the content of total flavonoids by spectrophotometry; we obtain the best extraction process.Secondly,we develop preparative HSCCC to isolate flavonoids compounds from ZSS. The solvent system is composed of ethyl acetate:n-butyl alcohol:water(3:2:5 v/v/v ), we can obtain three single compounds first time: spinosin,6'''-feruloylspinosin and derivative, The structures of compounds are identified by ESI-MS and papers, we develop a method for quantitative determination of spinosin, 6'''-feruloylspinosin and its derivative from ZSS carried out by HPLC.The third, The antioxidant potency of various solvents and concentration from ZSS is evaluated using three medthods, which scavenging·OH, O2-·, H2O2 produced from Fenton reaction, Pyrogallol self-oxidation system, Hydrogen peroxide system, specifically. The results show that the component of flavonoids from ZSS have a certain effect on scavenging·OH, O2-·, H2O2.In particular, it is obvious that the component of flavonoids from ZSS is good at scavenging Hydroxyl radical. Scavenging rate is 64%,it is more three time than Vc.
