| Parkinson's disease(PD) is one of the most common neurodegenerative disorders which primarily affects the eldly population. With the aging of our population,the PD has been a seriously burden of our society and family life. It is essential to clarify its pathogenesis,to prevent and treat it,which could mitigate this disease and improve the quality of human's lives effectively.Many studies indicate that PD is caused by a variety of different reasons,but the finally performance is the substantia nigra dopaminergic neuronal apoptosis,that is,the eventual common pathway of PD's pathogenesis is apoptosis.This thesis mainly study about the effects of tremor-stopping capsule on the expression of apoptotic protein genes Caspase-3,Bcl-2 and Bax in the substantia nigra pars of PD's rats.Objective:On the base of establishing PD's rats model successfully,to explore the effects of tremor-stopping capsule on the expression of apoptotic protein genes Caspase-3,Bcl-2 and Bax in the substantia nigra pars of PD's rats.Method:Using the method of injecting 6 - hydroxydopamine (6-OHDA) in the substantia nigra pars of rat's brain located to get injured PD mode in part.We select normal SD rats as the experimental control group,and PD rats are divided into PD model group, tremor-stopping 7 days and 14 days. Reverse transcription polymerase chain reaction (RT-PCR) were used to the four groups all the above to analyse the expression of Caspase-3,Bcl-2 and Bax.The results of each group were statistically analyzed by SPSS,the groups are compared with each other to determine the effects of tremor-stopping capsule on the expression of apoptotic protein genes Caspase-3,Bcl-2and Bax in the substantia nigra pars of PD's rats .Results :â‘ The expression levels of Caspase-3 and Bax are increased in the PD model group compared with the control group,while Bcl-2 decreased.Both groups are analyzed by the SPSS,the reaslts are P<0.01 and the differences are significant.â‘¡The expression levels of Caspase-3 and Bax of the PD rats treated by tremor-stopping capsule are decreased compared with the PD model group,while Bcl-2 increased. All groups are analyzed by the SPSS,the reaslts are P<0.01 and the differences are significant.â‘¢The expression levels of Caspase-3 and Bax of the PD rats treated for 14 days by tremor-stopping capsule are decreased compared with the traeted for 7 days group,while Bcl-2 increased.Both groups are analyzed by the SPSS,the reaslts are P <0.01 and the differences are significant.Conclusions:The expression levels of Caspase-3 and Bax in the substantia nigra pars of SD's rats are increased caused by PD, while it is decreased of Bcl-2.The PD's rats who treated by the tremor-stopping capsule their expressions of Caspase-3 and Bax are decreased,while Bcl-2 increased.And as the time of treatment gose on, the effects are more obvious.It is indicated that the traditional Chinese medicine tremor-stopping capsule has some inhibition on the apoptosis of the substantia nigra pars in PD's rats. |
