Objective:(1)Corneas of BALB/C and C57BL/6 are infected with Fusarium solani and Aspergillus fumigatus. (2) After infection, we observe the clinical process; (3)After infection,we detect the level change of Thl-related IFN-Y and Th2-related IL-4.Methods:(1)Infection of mice:epitheliums and superficial stroma of central corneas are eliminated and wounded by stroma-incision hook.5μl Suspension of Fusarium solani or Aspergillus fumigatus are applied to the eye surface.(2)Slit-lamp microscope are used to observe the clinical process of fungal keratitis for successive 7 days.(3)Infected eyes and ipsilateral cervical lymphatic nodes are enucleated homogenized after infection at 24h,48h and 72h.We use Elisa to detect Thl-related IFN-γand Th2-related IL-4.Results:(1)Fungal keratitis of BALB/C recover within 7 days, with slight or moderate edema and inflammatory infiltration in the process, leaving no or slight opacity. By contrast, Fungal keratitis of C57BL/6 is much more severe in edema and inflammatory infiltration and perforation occurs after 3 days, often culminating in dense opacity or even shrink.(2)After BALB/C mice are infected with Fusarium solani or Aspergillus fumigatus, the level of IFN-Y elevates both in eyes and lymphatic nodes. The peak of level in eyes is 2.9 times as uninfected level for Fusarium solani and 2.6 times for Aspergillus fumigatus. The peak of level in lymphatic nodes is 2.0 times as uninfected level for Fusarium solani and 1.5 times for Aspergillus fumigatus. For C57BL/6, the level of IFN-γonly slightly elevates in eyes, the peak is 1.3 times for Fusarium solani and 1.1 times for Aspergillus fumigatus.The peak in lymphatic nodes is 1.6 times and 1.5 times.(3) After BALB/C mice are infected with Fusarium solani or Aspergillus fumigatus, the level of IL-4 in eyes reduced, the trough is 0.6 times as uninfected level for Fusarium solani and 0.5 times for Aspergillus fumigatus. But the level in lymphatic nodes elevates,with the peak 1.2 times and 1.5 times. Similarly for C57BL/6,the trough of the level in eyes is 0.6 times and 0.5 times, with the peak in lymphatic nodes 1.9 times and 1.6 times.Conclusions:(1)Thl-type adaptive immune response is induced by fungal keratitis;(2)The level of local Thl-polarizaion is crucial to prognosis; (3) The level of local Thl-polarizaion of BALB/C is higher than C57BL/6;(4)In the process of fungal keratitis, infectious location and lymphatic nodes are equidirectional in Th-1 polarization but reverse in Th-2 polarization.
