ObjectiveTo research the COX-2/PGE2/P450arom signaling pathway mediated by HER-2/neu in the action of estrogen-dependent endometrial cancer, transfect the HER-2/neu into human endometrial carcinoma Ishikawa cells, detect the difference in the expressions of COX-2,P450arom and E2 between the transfected and non-transfected cell lines.Methods1. Culture the human endometrial carcinoma Ishikawa cells;2. Transfect the HER-2/neu into human endometrial carcinoma Ishikawa cells, build over-expressed HER-2/neu cell line;3. The difference was detected in the expressions of COX-2, P450arom between the transfected and non-transfected cell lines by Western blot;4. The difference was detected in the expressions of E2 between the transfected and non-transfected cell lines by ELISA;5. Analyze the experimental results by SPSS software, then analysis the action of HER-2/neu in regulation of COX-2/PGE2/P450arom signaling pathway, discussing the relationship between HER-2/neu and estrogen-dependent endometrial cancer.Results1. establish a stable transfection of HER-2/neu gene in human endometrial adenocarcinoma Ishikawa cells, the expression of HER-2/neu is increased(P<0.05);2. The expression of HER-2/neu is increased(P<0.05), meanwhile the expression of COX-2 and P450arom protein is also increased significantly in the transfected cell lines(P<0.05); 3. The expression of HER-2/neu,COX-2 and P450arom is increased(P<0.05) meanwhile the expression of E2 is also increased significantly in the transfected cell lines(P<0.05);The expression of is been positively correlated with the expression of COX-2, P450arom protein.ConclusionsThe over-expression of HER-2/neu in Ishikawa endometrial cancer cell lines may regulate the COX-2/PGE2/P450arom signaling pathway, which cause that the expression of E2 increased.
